摘要：目的探讨益气消癥方对子宫肌瘤大鼠血液流变学和免疫功能的影响。方法参照雌孕激素联合诱导法建模,随机分成:空白对照(Control)组、模型对照(Model)组、桂枝茯苓胶囊(G-F)组、益气消癥方低(Y-L)、中(Y-M)、高(Y-H)剂量组。造模成功后各组按对应剂量灌胃干预,连续30 d。计算子宫系数,观测子宫病理形态学改变和检测ER和PR的含量、血液流变学(血浆黏度、红细胞比容及不同切变下的全血黏度)及免疫功能(CD_(3)^(+)、CD_(4)^(+)、CD_(8)^(+))。结果光镜下Model组可见明显增厚的子宫平滑肌,以空泡、大小不等的细胞、排列紊乱的肌组织、增生活跃的腺体呈现;其中子宫系数、ER和PR、血浆黏度及红细胞比容、CD_(8)^(+)水平:Control组与Model组、Y-H组与G-F组、Y-H组与Y-L组、Y-M组后者较前者均有不同程度的增高;而CD_(3)^(+)、CD_(4)^(+)的水平后者显著降低(P<0.05)。结论益气消癥方对子宫肌瘤大鼠有显著的干预效应,其机制与调控血液流变学、提高机体免疫功能、抑制靶细胞对雌孕激素的反应性密切相关。
Objective To investigate the effect of Yiqi Xiaozheng Recipe(益气消癥方)on hemorheology and immune function of uterine fibroids in rats.Methods The combined induction method of estrogen and progesterone was used and the model rats were randomly divided intoblank controlgroup,model control group,Guizhi Fuling Capsule(桂枝茯苓丸,G-F)group,Yiqi Xiaozheng Recipe low(Y-L),medium(Y-M)and high(Y-H)dose groups.After successful modeling,each group was given the corresponding doses of intragastric intervention for 30 consecutive days.The uterine coefficient was calculated,the pathological changes of the uterus were observed and the contents of ER and PR,hemorheology(plasma viscosity,hematocrit and whole blood viscosity under different shearing conditions)and immune function(CD+3,CD+4,CD+8)were detected.Results Under the light microscope,the uterine smooth muscle was obviously thickened in the model group,presented with vacuoles,cells of different sizes,disordered muscle tissueand active glands.The uterine coefficient,the contents of ER and PR,plasma viscosity and hematocrit and CD8+level:control group VS model group,Y-H group VS G-F group,Y-H group VS Y-L group and Y-M group,those of the latter groups were all increased to varying degrees compared with those the former groups;while the levels of CD+3and CD+4were significantly decreased in the latter groups(P<0.05).Conclusion Yiqi Xiaozheng Recipe has a significant intervention effect on uterine fibroids in rats,and its mechanism is closely related to regulatinghemorheology,improving immune functionand inhibiting the responsiveness of target cells to estrogen and progesterone.
