摘要:目的探讨贞芪扶正颗粒辅助奥沙利铂治疗H22移植瘤小鼠的疗效及其对细胞凋亡相关因子B淋巴细胞瘤?2(Bcl?2)和Bcl?2相关X的蛋白质(Bax)的影响。方法腹腔驯化H22细胞悬液,于前肢右侧腋下接种建立KM小鼠H22荷瘤模型,以奥沙利铂+贞芪扶正颗粒(高、中、低剂量)干预10 d,并进行常规观察、称重和检测瘤体变化,末次给药24 h后处死小鼠,测量瘤体大小和计算抑瘤率及脾脏指数,应用免疫组化法和qPCR分别检测瘤体组织中Bcl?2、Bax的蛋白和mRNA表达、全自动蛋白电泳分析法(Simple Western)测定Bcl?2、Bax蛋白表达。结果与模型组比较,各药物组小鼠体质量均减轻(P<0.05),瘤体体积减小、质量下降(P<0.05);与奥沙利铂组比较,贞芪扶正颗粒(高、中、低剂量)+奥沙利铂组抑瘤率均增高(P<0.05),脾脏质量与脾脏指数减小(P<0.05),Bcl?2蛋白和mRNA降低(P<0.05),Bax蛋白和mRNA升高(P<0.05)。结论贞芪扶正颗粒具有增强奥沙利铂抑制肝癌移植瘤生长的作用,其分子机制可能与调节Bcl?2和Bax的表达有关。
AIM To explore the curative effect of Zhenqi Fuzheng Granules assisted with oxaliplatin on micetransplanted with H22tumor cell and the influence on apoptosis related factors Bcl?2 and Bax levels existing in tumor tissue.METHODS H22?bearing mice model was established by injection of celiac domestication H22 cell in the right armpit of KM mice.The mice were observed daily after intraperitoneal injection of oxaliplatin and gavage of zhenqifuxia granules(high,medium and low dose)for 10 days and the tumor body was weighed and measured.After last dosing 24 h,the mice were killed by dislocation,and the tumor sige was measured and the spleen index and inhibitory rate were calculated.The protein and mRNA levels of Bcl?2 and Bax in tumor tissues were detected by methods of IHC,WB and qPCR.RESULTS Compared with the model group,the weight of mice and the tumor volume in each treatment group decreased(P<0.05).Compared with the oxaliplatin group,the tumor inhibition rate(P<0.05)in Zhenqi Fuzheng Granules(high,medium and low dose)+oxaliplatin groups increased(P<0.05),the spleen quality and the spleen index decreased(P<0.05),the protein and mRNA levels of Bcl?2 decreased(P<0.05),but the levels of Bax increased(P<0.05).CONCLUSION Zhenqi Fuzheng Granules can enhance the inhibition of oxaliplatin on the growth of transplanted hepatoma and its molecular mechanism may be related to the expression of bcl?2 and Bax.
