摘要:目的:将黄连解毒汤全汤、上清液和沉淀作用于阿尔茨海默症(AD)模型大鼠,探究黄连解毒汤各部位对AD模型大鼠的治疗效果及作用机制。方法:选用42只SD大鼠构建AD大鼠模型,随机分为假手术组、模型组、阳性组(石杉碱甲,2×10^(-5)g/kg)、黄连解毒汤全汤组、上清液组和沉淀组,给药剂量为3 g/kg,灌胃28 d。采用水迷宫实验测试大鼠行为学能力,苏木精-伊红(HE)染色观察海马病理改变,酶联免疫吸附测定法(ELISA)检测血清和脑脊液中白细胞介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)水平。结果:与模型组相比,黄连解毒汤各治疗组逃避潜伏期均有不同程度的缩短(P<0.01),有效区域运动时间和距离及有效区域进入次数均增加(P<0.05,P<0.01)。病理结果表明模型组海马组织有较多锥体细胞坏死,胞核固缩,着色加深;黄连解毒汤各治疗组均出现少量锥体细胞坏死,且锥体细胞数目略有减少。与模型组相比,黄连解毒汤各部位组血清和脑脊液中IL-1β、TNF-α浓度均有不同程度降低(P<0.05,P<0.01)。与黄连解毒汤上清液组和沉淀组相比,黄连解毒汤全汤组中IL-1β、TNF-α浓度降低最多。结论:黄连解毒汤全汤通过其上清液和沉淀协同发挥治疗AD的作用,其主要机制是降低AD大鼠血清和脑脊液中IL-1β、TNF-α水平,减轻AD大鼠“外周-中枢”炎症,提高AD大鼠记忆能力。
Objective:To investigate the therapeutic effects and mechanism of action of the whole decoction,supernatant,and sedimentation of Huanglian Jiedu Decoction(HLJDD)on Alzheimer’s disease(AD)model rats by applying them to AD model rats.Methods:42 Sprague-Dawley(SD)rats were utilized to establish the AD rat model and were randomly assigned to six groups:the sham operation group,the model group,the positive control group(Huperzine A at a dose of 2×10^(-5)g/kg),the HLJDD whole formula group,the HLJDD supernatant group,and the HLJDD sedimentation group.The administration dosage was set at 3 g/kg,and oral gavage was performed daily for 28 consecutive days.The water maze was employed to evaluate the behavioral abilities of rats.Hematoxylin-eosin(HE)staining was employed to examine the pathological changes in the hippocampus.Enzyme-linked immunosorbent assay(ELISA)was conducted to measure the levels of interleukin-1β(IL-1β)and tumor necrosis factor-α(TNF-α)in both serum and cerebrospinal fluid.Results:All HLJDD-treated groups exhibited a significant decrease in escape latency(P<0.01)and demonstrated increased time spent as well as distance traveled in the target quadrant compared with the model group(P<0.05,P<0.01).The pathological results indicated that there were more pyramidal cell necrosis in the hippocampal tissue of the model group,with nucleus condensation and deepened staining.In the HLJDD treatment group,a small amount of pyramidal cell necrosis occurred and the number of pyramidal cells slightly decreased.Compared with the model group,the concentrations of IL-1βand TNF-αin serum and cerebrospinal fluid were reduced to different degrees in each treatment group of HLJDD(P<0.05,P<0.01).The concentrations of IL-1βand TNF-αwere most reduced in the HLJDD whole formula group compared with the HLJDD supernatant and HLJDD sedimentation groups.Conclusion:HLJDD whole decoction exerts its therapeutic effect on AD through the synergy of its supernatant and sedimentation.Its main mechanism is to reduce the levels of IL-1βand TNF-αin the serum and cerebrospinal fluid of AD rats,alleviate the“peripheral-central”inflammation of AD rats,and improve the memory ability of AD rats.
