摘要:目的:探讨古墨膏的抗炎作用及其作用机制。方法:采用小鼠耳肿胀、大鼠足跖肿胀及小鼠腹腔毛细血管通透性增高等模型观察古墨膏的抗炎作用;同时测定古墨膏对角叉菜胶致足炎性渗出物中的前列腺素E2(PGE2)、丙二醛(MDA)蛋白含量和超氧化歧化酶(SOD)活力的影响,以及小鼠气囊炎性渗出液白细胞数、一氧化氮(NO)含量。结果:古墨膏能显著抑制二甲苯所致小鼠耳肿胀(P<0.01),明显减轻大鼠足跖肿胀度(P<0.01)、降低足炎性渗出物中的PGE2、MDA、蛋白含量(P<0.01),显著增加足跖局部炎症组织中SOD的活性(P<0.01)、减少小鼠气囊渗出液中白细胞数和NO含量(P<0.01),并具有抑制醋酸引起的小鼠腹腔毛细血管通透性增加作用。结论:古墨膏具有明显的抗炎作用,其机制可能与抑制PGE2、NO等炎症介质合成、抑制脂质过氧化、增强自由基清除能力,抑制白细胞渗出有关。
Objective: To study the anti-inflammatory effects of Gumo cream and its mechanisms.Methods: The models of ear swelling in mice,paw edema in rats and increased capillary permeability in mice were established to observe the anti-inflammatory effect of Gumo cream.Meanwhile,the effects of Gumo cream on protein contents of prostaglandin E2(PGE2) and malondialdehyde(MDA) and the activity of superoxide dismutase(SOD) in carrageenan-induced inflammatory exudates as well as the white blood cell count and nitric oxide(NO) levels in the balloon inflammatory exudates in mice were detected.Results: Gumo cream can significantly inhibit xylene-induced ear swelling(P0.01),alleviate paw swelling in rats(P0.01),reduce the protein content of PGE2,MDA in foot inflammatory exudates(P0.01),increase the activity of SOD in plantar inflammatory tissue(P0.01),reduce the white blood cell count and nitric oxide(NO) levels in the balloon inflammatory exudates(P0.01),and inhibit the increase of peritoneal capillary permeability induced by acetic acid.Conclusion: Gumo cream has marked anti-inflammatory effects which may related to its inhibiting the synthesis of PGE2 and NO,reducing the peroxidation of lipid,increasing free radical scavenging capacity and inhibiting leukocyte exudation.
