摘要:目的:探讨白及多糖对胃溃疡(GU)模型大鼠胃组织JNK、p38 MAPK基因、蛋白及其介导炎性因子TNF-α、IL-1β和IL-6表达水平的影响。方法:将60只SPF级Wistar大鼠随机分为空白组和模型组,模型组大鼠采用乙酸直接烧灼法复制GU模型,将GU模型大鼠按随机数字表法分为模型组、盐酸雷尼替丁0. 3 g/kg组和白及多糖0. 125、0. 25、0. 5 g/kg组,每组10只。各组灌胃相应药物或蒸馏水,连续15日。采用ELISA法检测各组大鼠胃溃疡病灶组织炎症因子TNF-α、IL-1β及IL-6含量,RT-PCR法检测胃溃疡病灶组织TNF-α、IL-1β、IL-6、JNK及p38 MAPK基因表达水平,WB法检测JNK及p38 MAPK蛋白表达水平。结果:与空白组比较,GU大鼠病理见胃底腺结构紊乱,胃黏膜上皮细胞脱落等,溃疡长度显著增加,胃组织炎症因子TNF-α、IL-1β及IL-6含量升高,基因表达显著上调,JNK及p38 MAPK基因及蛋白表达水平显著上调;与GU模型组比较,白及多糖0. 5 g/kg组大鼠病理见胃黏膜出现较多腺体,炎症细胞浸润、坏死层明显减少,组织病理学检查评分明显降低;白及多糖各剂量组TNF-α、IL-1β及IL-6含量明显降低,基因表达均下调,JNK及p38 MAPK基因及蛋白表达水平均下调,以白及多糖0. 5 g/kg组最显著。结论:白及多糖可通过下调JNK及P38 MAPK基因和蛋白表达水平,抑制炎症因子TNF-α、IL-1β及IL-6异常分泌而发挥保护胃黏膜的作用。
Objective: To investigate the effects of Bletilla baicalensis polysaccharide on the expression of JNK,p38 MAPK gene protein and inflammatory factors TNF-alpha,IL-1 beta and IL-6 in gastric tissue of rats with gastric ulcer (GU). Methods: 60 SPF Wistar rats were randomly divided into the blank group and the model group. The GU model was replicated by direct acetic acid cauterization. After modeled,rats in the model group were randomly divided into five groups again: the model group,0. 3 g/kg ranitidine hydrochloride group and Bletilla baicalensis polysaccharide groups (0. 125、0. 25、0. 5 g/kg),10 rats in each group. Rats were administered with corresponding drugs or distilled water for 15 days. The levels of inflammatory factors TNF α、IL 1β and IL-6 in gastric ulcer tissues were detected by ELISA. The expression levels of TNFα,IL-1β,IL-6,JNK and p38 MAPK in gastric ulcer tissues were detected by RT-PCR. The protein levels of JNK and p38 MAPK were detected by Western blotting (WB). Results: Compared with the blank group,pathological findings of GU rats were observed,including gastric fundus gland structure disorder,exfoliation of gastric mucosal epithelial cells,significant increase of ulcer length,significant increase of inflammatory factors contents of TNF α、IL 1βand IL-6 and expression of mRNA,and significant increase of JNK and p38 MAPK expression levels. Compared with the model group,the pathological changes of rats in 0. 5 g/kg Bletilla baicalensis polysaccharide group showed that more glands appeared in gastric mucosa,inflammatory cells infiltrated and necrosis layer significantly decreased. In all doses of Bletilla baicalensis polysaccharide groups,the levels and expressions of TNF α、IL 1β and IL-6 were significantly decreased. The expression levels of JNK and p38 MAPK were decreased,while those in 0. 5 g/kg Bletilla baicalensis polysaccharide group showed most significant difference. Conclusion: Bletilla baicalensis polysaccharide can protect gastric mucosa by down-regulating gene and protein levels of JNK and P38 MAPK,inhibiting the abnormal secretion of inflammatory factors TNF α、IL 1βand IL-6.
