摘要:目的:研究分析牛蒡子-甘草药对治疗咽炎的潜在活性成分群及可能作用机制,并建立同时检测活性成分群中多种成分的HPLC条件与方法,为阐明其药效物质基础及分子机制的实验研究提供基础。方法:通过文献数据挖掘对牛蒡子-甘草药对治疗咽炎的潜在活性成分群进行初步筛选;运用网络药理学对筛选得到的活性成分进行分析;采用HPLC建立同时检测牛蒡子-甘草药对中13个活性成分的定量分析方法。结果:筛选得到由13个主要化学成分组成的牛蒡子-甘草抗咽炎潜在活性成分群;“成分-靶点”网络构建、PPI网络构建、GO功能分析与KEGG通路分析等的综合研究结果显示,牛蒡子-甘草药对抗咽炎可能性作用机制为:以甘草酸、异甘草素、异甘草苷、牛蒡苷元、牛蒡子苷、绿原酸等为主的活性成分群通过与STAT3、MAPK1、RELA、VEGFA等蛋白靶点结合,可调控癌症通路、T细胞受体信号通路、PI3K/AKT信号通路等多条与炎性反应和免疫调节密切相关的信号通路发挥作用。结论:筛选得到牛蒡子-甘草药对抗咽炎的药效物质基础是由13个主要成分组成的活性成分群;通过“多成分-多靶点-多通路”的潜在作用机制,达到调节机体免疫应答,抗炎、抗病原微生物(驱邪外出),镇咳祛痰、解热镇痛(解除表证)的协同作用而治疗咽炎;牛蒡子-甘草药对配伍有利于牛蒡子中活性成分的溶出。本研究将为基于药代动力学的牛蒡子-甘草药对配伍后的抗咽炎药效物质基础研究、作用机制研究、相关制剂的质量控制提供方向指导、基础依据及检测技术方法。
Objective: To study and analyze the potential active components group and possible mechanism ofFructus arctii-Glycyrrhiza uralensis pair in the treatment of pharyngitis, and to establish the HPLC conditions and methods for the simultaneous detection of multiple components in the active components group, so as to provide a basis for the experimental study on the material basis and molecular mechanism of its efficacy. Methods: The active components of Fructus arctii-Glycyrrhiza uralensis pair were screened by literature integration.The network pharmacology was used to construct the "component-target-pathway" network and analyze the GO function and KEGG pathway of the selected active components. And the HPLC method was used to establish a quantitative analysis method for simultaneous detection of 13 active components in Fructus arctii-Glycyrrhiza uralensis pair. Results: A group of potentially active components composed of 13 main chemical constituents were screened of Fructus arctii-Glycyrrhiza uralensis pair in the treatment of pharyngitis.The comprehensive research results of “component-target” network construction, PPI network construction, GO function analysis and KEGG pathway analysis showed that the possible mechanism of action of Fructus arctii-Glycyrrhiza uralensis pair against pharyngitis was that active components such as glycyrrhizic acid, isoliquiritin, isoliquiritigenin, arctiin, arctigenin, chlorogenic acid can regulate cancer pathway, T cell receptor signal pathway, PI3 K/AKT signal pathway and other signal pathways closely related to inflammatory response and immune regulation to produce effects by combining with STAT3, MAPK1, RELA, VEGFA and other protein targets. Conclusion: The active material basis of Fructus arctii-Glycyrrhiza uralensis against pharyngitis are a group of active components composed of 13 main components. Through the potential mechanism of multi-component, multi-target, multi-pathway, the treatment of pharyngitis can be achieved by regulating immune response, anti-inflammation, anti-pathogenic microorganism(exorcise out), antitussive and expectorant, antipyretic and analgesia(remove the apparent syndrome)through synergistic effect, Fructus arctii-Glycyrrhiza uralensis pairis conducive to the dissolution of active ingredients of Fructus arctii.This study provides direction guidance, basic basis and detection techniques for anti-pharyngitis material basic research, action mechanism research and quality control of related preparations based on pharmacokinetics of Fructus arctii-Glycyrrhiza uralensis pair against pharyngitis.
