摘要:目的通过生物信息学方法探讨KLHL7基因在胃癌与癌旁组织的差异表达及与胃癌临床病理特征的关系,探索KLHL7基因参与胃癌发生发展的机制。方法通过仙桃学术在线工具分析研究KLHL7基因在胃癌与癌旁组织间的差异表达及相关病理参数,利用在线工具cBioPortal和Sanger学术平台对与KLHL7具有相关性的共表达基因进行筛选,并开展京都基因和基因组百科全书(KEGG)信号通路富集分析。利用STRING和Cytoscape 3.8对KEGG聚类分析中发现的与肿瘤密切相关的基因进行关键核心基因筛选。结果KLHL7是胃癌中呈高表达的癌基因,与KLHL7基因具有相关性的共表达基因有2785个,包括上调2486个和下调299个,部分基因富集在与肿瘤关系密切的25个信号通路中,包括自噬、凋亡、细胞周期、慢性白血病、子宫内膜癌、肿瘤坏死因子(TNF)、缺氧诱导因子(HIF-1)、结肠癌和Wnt等信号通路。免疫细胞浸润分析发现,高表达的KLHL7与中央记忆型T细胞(Tcm)和辅助性T细胞(Th细胞)浸润呈正相关,与浆细胞样树突状细胞(pDC)和Th17细胞浸润呈负相关。经蛋白互作(PPI)网络分析结合Cytoscape 3.8分析筛选发现,其中获得相关性较强的4个共表达基因BBS10、PPP1CB、ARL13B和MEAF6是影响KLHL7生物学表型的关键核心基因。结论KLHL7基因是胃癌中潜在的癌基因,与胃癌侵袭转移、细胞免疫浸润和预后有关。共表达相关基因BBS10、PPP1CB、ARL13B和MEAF6是影响KLHL7生物学表型的关键核心基因。
Objective To investigate the differential expression of KLHL7 gene in gastric cancer and adjacent tissues and its relationship with clinicopathological features of gastric cancer,and to explore the mechanism of KLHL7 gene′s involvement in the occurrence and development of gastric cancer.Methods The differential expression and relevant pathological parameters of KLHL7 gene between gastric cancer and adjacent tissues were analyzed by Xiantao academic online tool,and the co-expression genes related to KLHL7 were screened by online tools cBioPortal and Sanger data platform,and the enrichment analysis of KEGG signal pathway was carried out.The key core genes identified by KEGG enrichment analysis and closely related to tumors were screened using STRING and Cytoscape 3.8.Results KLHL7 was a highly expressed oncogene in gastric cancer.There were 2785 co-expressed genes associated with KLHL7 gene,including 2486 up-regulated and 299 down-regulated.Some genes were enriched in 25 signaling pathways closely related to tumor,including autophagy,apoptosis,cell cycle,chronic leukemia,endometrial cancer,TNF,HIF-1,colon cancer,and Wnt signaling pathways.The immunocyte infiltration analysis showed that the overexpression of KLHL7 positively correlated with Tcm cells(Tcm)and T helper cells(Th cells)invasion,and negatively correlated with pDC and Th17 cell invasion.According to the protein-protein interaction(PPI)analysis and screening of Cytoscape 3.8,co-expressed genes BBS10,PPP1CB,ARL13B and MEAF6,which strongly correlated with KLHL7,were the key core genes affecting the biological phenotype of KLHL7.Conclusion KLHL7 gene is a potential oncogene in gastric cancer,which is related to invasion and metastasis,cellular immune infiltration and prognosis of gastric cancer.Co-expressed genes BBS10,PPP1CB,ARL13B and MEAF6 are the key core genes that affect the biological phenotype of KLHL7.
