摘要:目的通过研究三黄凝胶作用于大鼠耳廓痤疮模型时核因子(NF)-κB信号通路和抗炎细胞因子白细胞介素(IL)-37的表达水平,探究三黄凝胶治疗痤疮的作用机理。方法选择60只6~8周Wistar大鼠,采用随机抽样法分6组:空白组和模型组、阳性对照组以及三黄凝胶的低浓度、中浓度和高浓度组,每组10只大鼠,除空白组外,剩余各组在鼠的右侧耳廓开口处涂抹100%油酸来制作粉刺模型,持续涂药3周,在肉眼、病理学观察提示造模成功后开始外用药物。不使用药物于空白组和模型组,不同浓度的三黄凝胶则分别外抹于三黄凝胶低浓度和中浓度、高浓度组,使用0.025%浓度的维A酸乳膏外抹于阳性对照组,持续使用药物4周后,以酶联免疫吸附测定(ELISA)法检测每组NF-κB和抗炎细胞因子IL-37活性表达水平。结果所有治疗组的NF-κB和抗炎细胞因子IL-37活性表达水平都比模型组的表达水平低很多,差异有统计学意义(P<0.05);三黄凝胶的低浓度、中浓度组和阳性对照组比较差异无统计学意义(P>0.05);三黄凝胶高浓度组和阳性对照组比较差异有统计学意义(P<0.05)。结论三黄凝胶在治疗痤疮中的作用机制与降低NF-κB和抗炎细胞因子IL-37活性表达水平密切相关。
Objective In order to study the effects of Sanhuang gel on the expression of nuclear factor(NF)-κB signal pathway and anti-inflammatory cytokine interleukin(IL)-37 in the rats models of auricular acne.Methods Sixty wistar rats of six to eight weeks were randomly divided into six groups:blank group,model group and positive control group;and low,medium and high concentration groups of Sanhuang gel,ten rats in each group.Except blank group,the remaining groups were smeared with 100%oleic acid at the opening of right auricle to make acne models,which lasted for three weeks.Macroscopic and pathological observation suggested that the models were established after work,start external use of drugs.No drug was used in the blank group and model group,different concentrations of Sanhuang gel were applied to the low concentration,medium concentration and high concentration groups of Sanhuang gel,and 0.025%concentration of vitamin A cream was applied to the positive control group.After continuous use of the drug for four weeks,the activity of NF-κB and anti-inflammatory cytokine IL-37 in each group was detected by enzyme-linked immunosorbent assay(ELISA).Results The expression levels of NF-κB and anti-inflammatory cytokines IL-37 in all treatment groups were significantly lower than those in the model group(P<0.05).There were no significant differences among the low and medium concentration groups and the positive control group(P>0.05).The differences between Sanhuang gel high concentration group and positive control group were statistically significant(P<0.05).Conclusion The mechanism of Sanhuang gel in the treatment of acne is closely related to the decreasing of NF-κB and anti-inflammatory cytokines IL-37.
