摘要:目的:探讨复方地黄汤对帕金森病(PD)小鼠行为学、神经递质和氧化应激指标的药理作用机制。方法:利用MPTP制作PD小鼠模型,选择复方地黄汤水煎液组和水煎醇提组灌胃观察对PD小鼠行为学、神经递质(DA、DOPAC、HVA)和氧化应激指标(MDA、GSH、GSH-Px、SOD)的影响。结果:与模型组和水煎醇提组比较,水煎液组可明显改善小鼠行为学的表现,提高神经递质(DA、DOPAC、HVA)的含量,提高氧化应激指标GSH、GSH-Px、SOD含量,降低MDA含量(P<0.01),且疗效优于水煎醇提组(P<0.01)。结论:复方地黄汤水煎液提取法工艺合理,疗效显著,从行为学、神经递质、氧化应激等指标来看,对PD小鼠疗效最佳。
Objective: To investigate the pharmacological mechanism of Fufang Dihuang Decoction on behavior, neurotransmitter and oxidative stress in mice with Parkinson’s disease(PD). Methods: The mouse model of PD using MPTP, select the water decoction group and alcohol extract of Fufang Dihuang Decoction group. Gastric lavage to observe its effect on the behavior of PD mice, neurotransmitter(DA, DOPAC, HVA) and oxidative stress index(MDA, GSH, GSH-Px, SOD). Results: Compared with the mice in model group and that of water decoction and alcohol extract group, water decoction of Fufang Dihuang Decoction obviously improved the mice behavior performance, increased the content of neurotransmitter(DA, DOPAC, HVA), and oxidative stress indexes of GSH, GSH-Px, SOD, decreased MDA(P<0.01), and the effect was better than the alcohol extract group(P<0.01). Conclusion: The extraction process of Fufang Dihuang Decoction is the best extraction process, because it is reasonable with significant effect. From the behavioral, neurotransmitter, oxidative stress and other indicators, it is the best effcet for PD mice.
