摘要:目的:建立祖师麻色谱指纹图谱,探究祖师麻抗炎、镇痛作用的谱效关系,为祖师麻抗炎、镇痛作用的物质基础研究提供参考。方法:采用高效液相色谱(HPLC)法建立祖师麻的指纹图谱,利用《中药色谱指纹图谱相似度评价系统(2012版)》进行色谱图相似度评价;与对照品比对确定7-羟基香豆素、木犀草素、芫花素、紫丁香苷、祖师麻甲素、7、8-二甲氧基香豆素在色谱指纹图谱中的位置。利用耳肿胀试验和冰醋酸扭体试验分别得到祖师麻的肿胀抑制率和疼痛抑制率,采用偏最小二乘回归分析法和灰色关联法分析祖师麻抗炎、镇痛作用的物质基础。结果:从祖师麻指纹图谱中共指认出19个共有峰。其中4号峰为7-羟基香豆素,9号峰为祖师麻甲素,11号峰为紫丁香苷,15号峰为7、8-二甲氧基香豆素,16号峰为木犀草素,19号峰为芫花素。偏最小二乘法分析表明,祖师麻发挥抗炎作用的物质是1、6、9、10、19号峰所代表的化合物,发挥镇痛作用的物质是1、7、9、12、16、17、18、19号峰所代表的化合物;灰色关联法分析表明,祖师麻发挥抗炎作用的物质是11和19号峰所代表的化合物,发挥镇痛作用的物质是8、9、11、14、18、19号峰所代表的化合物。结论:祖师麻发挥抗炎作用的主要物质基础为19号峰(芫花素),发挥镇痛作用的主要物质基础为9(祖师麻甲素)、19(芫花素)和18号峰化合物所代表的化合物。
Objective: To establish the chromatographic fingerprint of Daphnes Giraldii Cortex and explore the spectrum-effect relationship of anti-inflammatory and analgesic effects of Daphnes Giraldii Cortex to provide references for the research on the material basis of its anti-inflammatory and analgesic effects. Methods:High-performance liquid chromatography(HPLC) was used to establish the fingerprint of Daphnes Giraldii Cortex,and the similarity of the chromatogram was evaluated by Similarity Evaluation System for Chromatographic Fingerprint of Traditional Chinese Medicine(2012 edition). The positions of 7-hydroxycoumarin, luteolin, genkwanin, syringin, daphnetin, and 7-8-dimethoxycoumarin in chromatographic fingerprints were determined by comparison with reference substances. The swelling inhibition rate and pain inhibition rate of Daphnes Giraldii Cortex were detected by ear swelling test and acetic acid-induced writhing test, respectively. The material basis of anti-inflammatory and analgesic effects of Daphnes Giraldii Cortex were analyzed by partial least squares regression analysis and grey correlation method. Results:Nineteen common peaks were identified from the fingerprints of Daphnes Giraldii Cortex. Peaks 4,9,11,15,16,and 19 were identified as 7-hydroxycoumarin, daphnetin, syringin, 7-8-dimethoxycoumarin, luteolin, and genkwanin, respectively. Partial least squares analysis showed that the compounds with anti-inflammatory effect were those represented by peaks 1,6,9,10 and 19,and the compounds with analgesic effect were those represented by peaks 1,7,9,12,16,17,18,and 19. Grey correlation analysis showed that the compounds with the anti-inflammatory effect were those represented by peaks 11 and 19,while the compounds with the analgesic effect were those represented by peaks 8,9,11,14,18 and 19. Conclusion:The main active components with the anti-inflammatory effect in Daphnes Giraldii Cortex were those represented by peak 19,and the main active components with the analgesic effect were those represented by peaks 9(daphnetin),19(genkwanin) and 18.
