摘要:目的探讨红景天苷(SAL)对低氧诱导的大鼠心肌细胞凋亡及氧化应激损伤、炎症反应的影响。方法采用H9C2大鼠心肌细胞,实验分为常氧组(21%O2)和低氧组(2%O2),低氧组心肌细胞在专用低氧培养箱中先进行24 h的低氧处理,然后添加药物,之后继续在低氧条件下处理24 h。采用CCK-8试剂盒检测大鼠心肌细胞活力,通过检测乳酸脱氢酶(LDH)活性观察大鼠心肌细胞氧化应激损伤程度,采用酶联免疫吸附试验(ELISA)测定炎症因子白细胞介素(IL)-1β及IL-6水平,通过实时定量逆转录聚合酶链式反应(RT-qPCR)检测凋亡相关基因缺氧诱导因子1α(HIF-1α)、Capase3 mRNA相对表达量。结果与常氧对照组比较,大鼠心肌细胞经低氧诱导处理后,细胞活力明显下降,细胞内LDH活性及炎症因子IL-1β、IL-6水平明显升高,Caspase3、HIF-1αmRNA表达显著上调,差异均有统计学意义(P<0.05或P<0.01或P<0.001);20μmol/L SAL可明显升高大鼠心肌细胞活力,降低LDH活性及IL-1β、IL-6水平,下调Caspase3、HIF-1αmRNA表达,差异均有统计学意义(P<0.05或P<0.01或P<0.001)。结论SAL可以有效缓解低氧诱导的大鼠心肌细胞氧化应激损伤和炎症反应,抑制心肌细胞凋亡。
Objective To investigate the effects of salidroside(SAL)on hypoxia-induced apoptosis,oxidative stress injury and inflammatory response in rat cardiomyocytes.Methods H9C2 rat cardiomyocytes were used.The experiment was divided into normoxia group(21%O_2)and hypoxia group(2%O_2).Cardiomyocytes in the hypoxia group were first treated with hypoxia for 24 hours in a dedicated hypoxia incubator,then drugs were added,and then they were treated under hypoxia for another 24 hours.Cell viability was detected by CCK-8 kit,the degree of oxidative stress injury was observed by detecting lactate dehydrogenase(LDH)activity,the levels of inflammatory factors interleukin(IL)-1βand IL-6 were determined by enzyme-linked immunosorbent assay(ELISA),and the relative expression levels of apoptosis-related genes hypoxia-inducible factor 1α(HIF-1α)and Caspase3 mRNA were detected by real-time quantitative reverse transcription polymerase chain reaction(RT-qPCR).Results Compared with the normoxia group,after hypoxia induction,the cell viability of rat cardiomyocytes decreased significantly,the intracellular LDH activity and the levels of inflammatory factors IL-1βand IL-6 increased significantly,and the expression of Caspase3 and HIF-1αmRNA was significantly upregulated,with statistically significant differences(P<0.05 or P<0.01 or P<0.001);20μmol/L SAL could significantly increase the viability of cardiomyocytes,reduce LDH activity and the levels of IL-1βand IL-6,and reduce the expression of Caspase3 and HIF-1αmRNA,with statistically significant differences(P<0.05 or P<0.01 or P<0.001).Conclusion SAL can effectively alleviate hypoxia-induced oxidative stress injury and inflammatory response in rat cardiomyocytes and inhibit cardiomyocyte apoptosis.
