摘要:目的运用网络药理学技术研究分析菊花-麦冬药对治疗干眼的潜在机制和分子学途径。方法BATMAN-TCM数据库中以“Score cutoff≥40,P<0.05”作为条件筛选出菊花-麦冬的有效活性成分,采用人类基因数据库(GeneCards)和人类孟德尔数据库(OMIM)筛选出疾病靶点,使用Cytoscape 3.7.2软件构建“疾病-药对-化合物-靶点”调控网络。利用String软件构建交集靶点蛋白互作网络和条形图,分析蛋白的相互作用关系及核心靶点。使用clusterProfiler R包对共同靶点进行GO功能富集分析与KEGG通路富集分析,得到菊花-麦冬治疗干眼核心基因的生物功能和信号通路。结果在BATMAN-TCM平台检索到菊花-麦冬活性化合物46个,预测靶点291个,构建出“疾病-药对-化合物-靶点”网络后发现菊花-麦冬药对治疗干眼与171靶基因密切相关。GO功能富集分析与KEGG通路富集分析结果显示菊花-麦冬治疗干眼可能涉及的GO功能156个,信号通路64条。结论菊花-麦冬治疗干眼具有多分子、多靶点、多途径的调控作用,主要通过作用于CAT、TNF、CREB1、ESR1、COMT、ADK、NT5C2、NT5E、AR、PTGS、NR3C1等靶点而发挥生物学效应。
OBJECTIVE To study and analyze the potential mechanism and molecular approach of chrysanthemum-Ophiopogon japonicus to treat Xerophthalmia by using network pharmacology technology.METHODS The effective active ingredients of Chrysanthemum-Ophiopogon japonicus were selected from the BATMAN-TCM database under the condition of“Score cutoff≥40,P<0.05”,and screened out using the Human Gene Database(GeneCards)and Human Mendel Database(OMIM)For disease targets,use Cytoscape 3.7.2 software to construct a“disease-drug pair-compound-target”regulatory network.Use String software to build a cross-target protein interaction network and bar graph to analyze protein interactions and core targets.The cluster profiler R package was used to perform GO function enrichment analysis and KEGG pathway enrichment analysis on common targets to obtain the biological functions and signaling pathways of the core genes of chrysanthemum-Ophiopogon japonicus treatment of Xerophthalmia.RESULTS 46 chrysanthemum-Ophiopogon japonicus active compounds and 291 predicted targets were retrieved on the BATMAN-TCM platform.After constructing the“disease-drug pair-compound-target”network,it was found that chrysanthemum-Ophiopogon japonicus is closely related to 171 target genes in the treatment of Xerophthalmia.The enrichment analysis of GO function and the enrichment analysis of KEGG pathway showed that chrysanthemum-Ophiopogon japonicus treatment of Xerophthalmia may involve 156 GO functions and 64 signal pathways.CONCLUSIONS Chrysanthemum-Ophiopogon japonicus has a multimolecular,multi-target,and multi-path way regulation effect on Xerophthalmia,mainly by acting on CAT,TNF,CREB1,ESR1,COMT,ADK,NT5C2,NT5E,AR,PTGS,NR3C1 Target to exert biological effects.
