摘要:目的 :探讨三黄凝胶通过调控P38丝裂原活化蛋白激酶(P38 mitogen activated protein kinase,P38MAPK)/细胞外调节蛋白激酶(extracellular regulated protein kinases,ERK)信号通路干预痤疮的作用机制。方法:将48只SPF级Wistar大鼠分为空白组、模型组、维A酸乳膏组及三黄凝胶低、中、高剂量组。除空白组外,其余各组大鼠右耳均匀涂抹100%油酸,每日1次,进行痤疮造模。造模3周后维A酸乳膏组及三黄凝胶低、中、高剂量组分别涂抹相应药物治疗,每日1次,治疗4周;空白组、模型组涂抹生理盐水,每日1次,共4周。治疗前后观察痤疮鼠耳的病理变化;酶联免疫吸附试验(enzyme-linked immunosorbent assay,ELISA)法测定血清磷酸化-丝裂原活化蛋白激酶P38抗体(P-P38MAPK)与磷酸化-细胞外信号调节激酶(P-ERK)含量。结果:与模型组比较,维A酸乳膏组及三黄凝胶低、中、高剂量组鼠耳表皮及角质层变薄,毛囊及皮脂腺扩张、角栓、角化物质堆积、淋巴细胞浸润等病理变化均改善。各组痤疮鼠血清P-P38MAPK与P-ERK含量较空白组高,差异有统计学意义(P<0.01);与模型组相比,治疗后维A酸乳膏组及三黄凝胶低、中、高剂量组血清P-P38MAPK与P-ERK含量下降,差异有统计学意义(P<0.01)。结论:三黄凝胶能降低痤疮鼠血清P-P38MAPK及P-ERK含量,促进恢复痤疮模型大鼠表皮微生态环境的动态平衡,改善痤疮症状。
Objective:To discuss the mechanism of Sanhuang gel in the intervention of acne through regulating P38MAPK/ERK signaling pathway.Methods:All 48 SPF grade Wistar rats were allocated to the blank group,the model group,tretinoin cream group,low,moderate and high dose groups of Sanhuang gel.Except the blank group,the right ears of the rats in the rest groups were applied with 100% oleinic acid evenly for external use,once each day,to prepare acne models.After three weeks of modeling,tretinoin cream group,low,moderate and high dose groups of Sanhuang gel were treated with the corresponding medicine respectively,once each day,for four weeks;the blank group and the model group accepted physiological saline for external application,once each day,for four weeks.To observe pathological changes of the acne rats’ ears before and after the treatment,ELISA method was used to detect the contents of P-P38MAPK and P-ERK.Results:Compared with the model group,the epidermal and corneous layers became thinner,pathological changes of hair follicles and sebaceous glands expansion,keratotic plugging and materials accumulation,lymphocytes infiltration were improved in tretinoin cream group,low,moderate and high dose groups of Sanhuang gel.the contents of P-P38MAPK and P-ERK in different groups of acne rats were higher than these of the blank group,and the difference was statistically significant(P<0.01);compared with the model group,the contents of P-P38MAPK and P-ERK in tretinoin cream group,low,moderate and high dose groups of Sanhuang gel were decreased after the treatment,and the difference had statistical meaning(P<0.01).Conclusion:Sanhuang gel could reduce the contents of P-P38MAPK and P-ERK in the serum in acne rats,promote dynamic equilibrium in epidermal microecological environment of acne rats,and improve acne symptoms.
