摘要:目的:研究党参水提物对D-半乳糖致衰老小鼠胸腺和肾脏组织形态及Bax、VEGF表达的影响。方法:将100只昆明种小鼠随机分为对照组,衰老模型组,低、中、高剂量党参干预组。采用皮下注射D-半乳糖注射液的方法制备衰老模型后进行灌胃给药。计算各组小鼠的胸腺指数和肾脏指数,显微观察其组织结构,并采用免疫组织化学法检测胸腺及肾脏组织中Bax、VEGF的表达。结果:与对照组比较,衰老模型组小鼠胸腺指数和肾脏指数均明显下降(P<0.05);胸腺组织萎缩,免疫细胞减少,皮髓质分界不清,肾小球组织体积变大,细胞数量减少,内皮细胞肿胀;Bax、VEGF在胸腺和肾脏组织中阳性表达均明显增强(P<0.05)。与衰老模型组比较,高剂量党参干预组胸腺指数升高(P<0.05);胸腺皮质增厚,淋巴细胞密集,肾小球体积及细胞数目接近正常;Bax在胸腺和肾脏组织中阳性表达均明显减少(P<0.05),VEGF在肾脏组织中阳性表达明显增加(P<0.05)。结论:党参水提物对衰老小鼠胸腺和肾脏组织结构有明显的保护作用,其机制可能与抑制细胞凋亡、促进血管生成有关。
Objective :To investigate the effect of Codonopsis pilosula water extract on the morphology of the thymus and kidney and the expression of Bax and vascular endothelial growth factor(VEGF) in aging mice induced by D - galactose. Methods :A total of 100 Kunming mice were randomly divided into control group ,aging model group ,and low - , middle -, and high - dose Codonopsis pilosula intervention groups. Subcutaneously injectedD - galactose was used to establish an aging model and the drug was administered by gavage. The thymus index and renal index were calculated, a microscope was used to observe the structure of the thymus and kidney, and immunohistochemistry was used to measure the expression of Bax and VEGF in the thymus and kidney. Results: Compared with the control group,the aging model group had significant reductions in the thymus index and renal index ( P 〈 0. 05), as well as atrophy of the thymus, a reduction in immune cells, unclear boundary between the cortex and the medulla, an increased glomerular volume,a reduction in the number of cells, and endothelial swelling. There was a significant increase in the expression of Bax and VEGF in the thymus and kidney (P 〈 0. 05). Compared with the aging model group,the highdose Codonopsis pilosula intervention group had a significant increase in the thymus index(P 〈 0. 05),as well as thymic cortex thickening, an increase in lymphocytes, and nearly normal glomerular volume and number of cells;there was a significant reduction in the expression of Bax in the thymus and kidney(P 〈 0. 05)and a significant increase in the expression of VEGF in the kidney ( P 〈 0. 05 ). Conclusion : Codonopsis pilosula water extract can significantly protect the structure of the thymus and kidney in aging mice,possibly by inhibiting cell apoptosis and promoting angiogenesis.
