摘要:目的观察化浊颗粒对2型糖尿病(Type 2 diabetes mellitus,T2DM)大鼠肝组织微小RNA-21(microRNA-21,miR-21)/磷酸酶及张力蛋白同源基因(PTEN)/PI3K/AKT信号通路的调控作用。方法将40只SPF级雄性Wistar大鼠随机分为正常对照组、模型组、中药组、罗格列酮组。除正常对照组外,其余组别采用高糖、高脂饲料喂养联合小剂量链脲佐菌素(STZ)腹腔注射建立T2DM大鼠模型。造模后中药组及罗格列酮组分别以化浊颗粒和罗格列酮灌胃,模型组与正常对照组给予生理盐水灌胃。连续灌胃10周后处死各组大鼠,Q-PCR检测大鼠肝组织中miR-21、PTEN、PI3K、AKT mRNA水平,Western blot检测大鼠肝组织PTEN、PI3K、AKT、p-AKT蛋白表达。结果与正常对照组比较,模型组PTEN蛋白及mRNA表达下调,miR-21、PI3K、AKT mRNA表达水平升高,PI3K、AKT、p-AKT蛋白表达明显上调,差异有统计学意义(P<0.01);与模型组比较,中药组及罗格列酮组大鼠肝组织中PTEN蛋白及mRNA表达上调,miR-21、PI3K、AKT mRNA表达水平降低,PI3K、AKT、p-AKT蛋白表达明显下调,差异有统计学意义(P<0.05)。结论T2DM大鼠肝组织中miR-21及其靶基因PTEN调控的PI3K/AKT信号通路表达异常,化浊颗粒通过下调miR-21、上调PTEN表达,进而抑制PI3K/AKT信号通路、改善胰岛素抵抗,发挥治疗T2DM的作用。
Objective To observe theregulatory effect of HuazhuoGranules on the microRNA-21(miR-21)/PTEN/PI3 K/Aktsignaling pathway in liver tissues of rats with type 2 diabetes mellitus(T2 DM).Methods Forty SPF male Wistar rats were randomly divided into a normal control group(NCG),a model group(MG),a Chinese medicine group(CMG),and a rosiglitazone group(RSG).The T2 DM model was induced by intraperitoneal injection of streptozotocin(STZ)combined with high-sugar and high-fat diet in rats except for those in NCG.Rats in CMG and RSG were treated with HuazhuoGranules and rosiglitazone by gavage,respectively,and those in NCG and MG with normal saline.After treatment for 10 weeks,all rats were sacrificed.Q-PCR was used to detect the mRNA expression of miR-21,PTEN,PI3 K,and Akt in liver tissues of rats.Western blotwas used to detect the protein expression of PTEN,PI3 K,Akt,and p-Akt in liver tissues of rats.Results Compared with NCG,MG showed down-regulatedprotein and mRNA expressionof PTEN,up-regulatedmRNA expression of miR-21,PI3 K,and Akt,and increased protein expression of PI3 K,Akt,and p-Akt(P<0.01).Compared withMG,CMG and RSG showed up-regulatedprotein and mRNA expressionof PTEN,down-regulated mRNA expression of miR-21,PI3 K,and Akt,and reduced protein expression of PI3 K,Akt,and p-Akt(P<0.05).Conclusion The PI3 K/Akt signaling pathway regulated by miR-21 and its target gene PTEN is abnormally expressed in the liver tissues of T2 DM rats.Huazhuo Granules can treat T2 DM by down-regulating miR-21 expression and up-regulating PTENexpression,thereby inhibiting the PI3 K/Akt signaling pathway and improving insulin resistance.
