摘要:目的评价藏药二十五味儿茶凝胶治疗大鼠慢性湿疹的疗效。方法采用2,4-二硝基氯苯(DNCB)诱导大鼠背部慢性湿疹。将72只SD大鼠随机分为两组,取12只大鼠作为空白对照组,另一组共60只造模成功后随机分为模型组、阳性对照组、藏药二十五味儿茶凝胶高、中、低剂量组5组。阳性对照组给予丁酸氢化可的松乳膏(尤卓尔),藏药二十五味儿茶凝胶低(0.14g/kg)、中(0.21g/kg)、高(0.35g/kg)剂量经皮给药,于致敏后给药,观察各组大鼠第4次激发3天后采用ELISA法检测大鼠血清中的LTB-4、LTC-4的含量。结果藏药二十五味儿茶凝胶中剂量能有效减轻慢性湿疹大鼠引起的皮损炎症程度,调节慢性湿疹组大鼠血清中LTB-4和LTC-4水平。结论藏药二十五味儿茶凝胶能降低慢性湿疹模型大鼠血清LTB-4和LTC-4含量,其治疗湿疹的作用机制可能与免疫调节及减轻炎症反应相关。
Objective To evaluate the therapeutic effect of Tibetan drug Ershiwuweiercha gel on chronic eczema in model rats. Methods A total of 72 SD rats with chronic eczema induced by 2, 4-dinitrochlorobenzene (DNCB) were randomly divided into two groups: blank control group (n = 12), and the other group consisting of 60 success- fully model-made rats was randomly subdivided into model group, positive control group, high, medium and low dose of Tibetan drug Ershiwuweiercha gel group. Posi- tive control group was given hydrocortisone butyrate cream (Youzhuoer), low-dose (0.14g/kg), medium-dose (0.21g/kg), and high-dose (0.35g/kg) group were treated with percutaneously absorbed drugs after sensitization. Three days after obser- vation of the fourth challenge in each group, serum LTB-4 and LTC-4 levels were de- tected by ELISA. Results The medium-dose Tibetan drug Ershiwuweiercha gel was effective in the amelioration of skin inflammation caused by rats with chronic eczema, and as well as in the adjustment of the levels of ltb-4 and hc4 in the serum of rats with chronic eczema. Conclusion Serum ltb-4 and ltc-4 levels in rats with chronic eczema can be decreased by Tibetan drug Ershiwuweiercha gel, and its mechanism may be associated with immune regulation and reducing inflammatory response.
