摘要:microRNA在肌、骨代谢疾病中的作用已成为当前研究热点。本文对microRNA在肌少-骨质疏松症中的作用机制研究进行总结。首先,本文介绍microRNA的生物发生过程和基因调控机制。其次,本文分别介绍相关microRNA亚型在骨骼肌和骨骼重建中的作用。在microRNA对骨骼肌作用机制方面,归纳了5个方面:(1)调节转录因子MyoD的活化和表达;(2)调节其他肌肉特异性转录因子的表达;(3)影响肌肉细胞的增殖和分化;(4)影响肌肉细胞表型;(5)microRNA的协同作用。并从促进成骨和抑制破骨两方面说明microRNA影响骨重塑的作用机制。还对具有肌骨共生的microRNA进行了总结归纳。最后对相关研究中存在的问题与不足进行总结和展望,对肌少-骨质疏松症的防治具有积极意义。
The role of microRNA in muscle and bone metabolic diseases has become a research hotspot.This article summarized the research on the mechanism of action of microRNA in osteo sarcopenia.Firstly,this article introduced the biological process and gene regulation mechanism of microRNA.Secondly,this article introduced the roles of relevant microRNA subtypes in skeletal muscle and bone reconstruction.In terms of the mechanism of action of microRNA on skeletal muscle,five aspects had been summarized:(1)regulating the activation and expression of the transcription factor MyoD;(2)regulating the expression of other muscle specific transcription factors;(3)affecting the proliferation and differentiation of muscle cells;(4)affecting the phenotype of muscle cells;(5)the synergistic effect of microRNA.And the mechanism of microRNA affecting bone remodeling was explained from two aspects:promoting osteogenesis and inhibiting osteoclast.microRNAs involved in muscle and bone generation were also summarized.Finally,a summary of the problems and shortcomings in relevant research was of positive significance for the prevention and treatment of osteosarcopenia.
