摘要:目的:观察黄芪对心脏骤停后综合征(post-cardiac arrest syndrome,PCAS)模型家兔炎性因子的影响。方法:将清洁级家兔随机分为假手术组、PCAS组、肾上腺素+黄芪组、黄芪组,每组10只,通过室颤法建立兔PCAS模型,假手术组不造成窒息。于复苏前、复苏后6、24、48 h分别从意识、行为、转头、转圈及偏盲5方面进行兔神经损伤评分;复苏后24 h取股静脉血、复苏后48 h取兔脑组织用ELISA法检测血清及脑组织中炎性因子的表达。结果:1)与PCAS组比较,肾上腺素+黄芪组、黄芪组神经功能损伤评分在复苏后24、48小时减小;2)与假手术组比较,PCAS组血清白细胞介素1β(IL-1β)、白细胞介素8(IL-8)含量及脑组织IL-1β、IL-8含量均升高(P<0.05);3)与PCAS组比较,复苏后24 h肾上腺素+黄芪组、黄芪组血清及脑组织中IL-1β、IL-8含量减少,且肾上腺素+黄芪组较黄芪组减少更明显(P<0.05)。结论:肾上腺素联合黄芪能减轻家兔自主循环恢复后脑细胞损伤,减少复苏后脑组织炎性因子表达,改善神经功能,其机制可能与抑制细胞凋亡有关。
Objective:To survey the effects of Huangqi(astragali radix)on inflammatory factors in the rabbits with post-cardiac arrest syndrome(PCAS).Methods:The rabbits in the clean level were randomized into sham operation group,PCAS group,adrenaline+Huangqi group and Huangqi group,ten rabbits each group,PCAS rabbit models were established via ventricular fibrillation method,and sham operation group didn′t cause asphyxia.Animal nerve injury scores were performed from five aspects including the consciousness,behavior,turning head,circling and hemiopia before the recovery,in six hours,24 hours and 48 hours after the recovery;femoral venous blood was drawn within 24 hours after the recovery,the expressions of inflammatory factors in the serum and cerebral tissue were detected using ELISA method.Results:1)Compared with PCAS group,nerve injury scores of adrenaline+Huangqi group and Huangqi group reduced notably in 24 hours and 48 hours after the recovery;2)Compared with sham operation group,the levels of IL-1βand IL-8 in the serum and the contents of IL-1βand IL-8 in cerebral tissue of PCAS group rose significantly(P<0.05);3)Compared with PCAS group,the expressions of IL-1βand IL-8 in the serum and the contents of IL-1βand IL-8 in cerebral tissue of adrenaline+Huangqi group and Huangqi group lowered notably within 24 hours after the recovery,and adrenaline+Huangqi group was more notable than Huangqi group(P<0.05).Conclusion:Adrenaline combined with Huangqi could relieve brain cell damage after the restoration of spontaneous circulation,reduce the expressions of inflammatory factors in the cerebral tissue after the recovery and improve neurological function,its mechanism might be related to inhibiting cellular apoptosis.
