摘要:目的优化痛风片的提取工艺及其片剂的成型工艺。方法以加水倍数、煎煮时间、煎煮次数为影响因素,山栀苷甲酯、8-O-乙酰山栀苷甲酯的含量及含膏量为评价指标,正交实验结合信息熵权法优化最佳提取工艺;以辅料与浸膏粉比例、淀粉与微晶纤维素比例、崩解剂用量为影响因素,脆碎度、硬度、休止角和崩解时限为评价指标,星点设计-效应面法结合总评归一(OD)法优化成型工艺。结果最佳提取工艺为8倍量水煎煮3次,每次0.5h,山栀苷甲酯、8-O-乙酰山栀苷甲酯质量分数分别为1.0652mg/g、0.9859mg/g,干膏得率为21.26%,综合评分为99.12分;最佳成型工艺为:辅料与浸膏粉比例0.4:1,淀粉与微晶纤维素比例0.85:1,崩解剂用量为8%,脆碎度、硬度、休止角、崩解时限分别为0.51%,116.2N,39.5°,42min,OD值为0.7260。结论该实验优化痛风片提取工艺及成型工艺简单、稳定可行,为进一步研究开发提供了依据。
Objective To optimize the extraction and molding process of Tongfeng tablets.Methods The optimum extraction process was optimized by orthogonal experiment combined with information entropy weight method,with the influencing factors of water addition times,decocting time and decocting times,the content and paste content of shanzhiside methyl ester and 8-O-acetyl shanzhiside methyl ester as evaluation indexes;taking the ratio of auxiliary material to extract powder,the ratio of starch to microcrystalline cellulose and the dosage of disintegrating agent as influencing factors,brittleness,hardness,angle of repose and disintegration time limit as evaluation indexes,central composite design response surface method combined with overall desirability(OD)method was used to optimize the molding process.Results The optimum extraction process was 8 times of water decoction for 3 times,0.5h each time.The mass fractions of shanzhiside methyl ester and 8-O-acetyl shanzhiside methyl ester were 1.0652mg/g and 0.9859mg/g,respectively.The yield of dry ointment was 21.26%,and the comprehensive score was 99.12 points;the optimum molding process were as follows:the ratio of auxiliary material to extract powder was 0.4:1,the ratio of starch to microcrystalline cellulose was 0.85:1,the dosage of disintegrating agent was 8%,the brittleness,hardness,angle of repose and disintegration time were 0.51%,116.2N,39.5°,42min respectively,and the overall evaluation OD was 0.7260.Conclusion The experiment optimizes the extraction process and molding process of Tongfeng tablets,which is simple,stable and feasible,and provides a basis for further research and development.
