摘要:目的探讨中药红芪提取物红芪多糖(HPS)联合右美托咪定(DEX)在心气虚模型大鼠心肌缺血再灌注(I/R)损伤中抗氧化应激可能的分子机制。方法18月龄雄性SD大鼠60只采用随机数字表法分为4组,比较血清甘油三酯(TG)、总胆固醇(TC)、肌钙蛋白(cTn1)、肌酸激酶同工酶(CK-MB)水平,取心肌组织,比较心肌蛋白激酶C(PKC)、磷脂酰肌醇3激酶(PI3K)、核因子相关因子2(Nrf2)mRNA表达及心肌病理学改变。结果与假手术组比较,再灌注组血清TG、TC、cTn1、CK-MB水平明显升高(P<0.05);与模型组比较,DEX+HPS和DEX组血清TG、TC、cTn1、CK-MB水平明显下降(P<0.05)。与正常组比较,模型组PKC、PI3K mRNA表达升高,Nrf2 mRNA表达下降;与模型组比较,各治疗组PKC、PI3K mRNA表达下降,Nrf2 mRNA表达升高(P<0.05)。结论红芪多糖联合右美托咪定预处理通过PKC、PI3K、Nrf2减轻氧化应激从而改善大鼠心肌缺血再灌注损伤。
Objective To investigate the potential molecular mechanism underlying the anti-oxidative stress of hedysarum polybotys saccharide combined with dexmedetomidine(DEX+HPS)using the rat model of deficiency of vital energy.Methods 60 male SD rats aged 18 month were randomly divided into 4 groups.Serum was collected for measuring and comparing the levels of triglycerides,total cholesterol,cTn1 and CK-MB;myocardial tissue was collected for measuring mRNA levels of PKC,PI3 K,and Nrf2,and observing pathological changes.Results The serum levels of triglycerides,total cholesterol,cTn1 and CK-MB in model group were significantly increased(P<0.01)when compared with the normal group.The serum levels of triglycerides,total cholesterol,cTn1 and CK-MB in treating group were significantly decreased(P<0.05)as compared with the model group.When compared with the normal group,the mRNA levels of PKC and PI3K in model group were significantly increased in the model group,whereas the mRNA level of Nrf2 was significantly decreased.When compared with the model group,the mRNA levels of PKC and PI3K in treated group were significantly decreased,whereas the mRNA level of Nrf2 was significantly increased(P<0.05).Conclusion Astragalus polysaccharide combined with Dexmedetomidine preconditioning can alleviate oxidative stress through PKC,PI3K and NRF2 and improve myocardial ischemia-reperfusion injury in rats.
