摘要:目的:从全基因组甲基化的角度探讨辐射损伤大鼠的旁效应发生机制。方法:采用12C6+束照射大鼠右侧肺部构建辐射损伤动物模型,同时设立正常组,取心、肝、脾、左肺、左肾组织以提取DNA。采用MethylRAD技术进行全基因组甲基化检测。结果:与正常组比较,模型组大鼠心、肝、脾、左肺、左肾组织中的甲基化位点(CCGG/CCWGG位点数,CCGG/CCWGG平均深度)显著降低(P<0.01),甲基化位点测序深度显著降低(P<0.01)。两组大鼠基因组不同功能元件上的甲基化位点水平高低不同,差异有统计学意义(P<0.01)。从染色体图谱来看,两组大鼠差异甲基化位点在染色体上的分布是不均匀的。结论:全基因组甲基化差异可能是辐射损伤大鼠旁效应的发生机制之一。
Objective: To discuss the mechanism of bystander effect in radiation injured rats from the point of whole genome methylation. Methods: Radiation injured rats model was established by 12 C6+ beam irradiated the right lung of rats. Meanwhile, a normal control group was set up, rats were sacrificed and DNA was extracted from heart, liver, spleen, left lung and left kidney. The whole genome methylation was detected by MethylRAD technique. Results: Compared with the normal group, methylation sites in heart, liver, spleen, left lung, left kidney tissues of model rats(CCGG/CCWGG loci, the average depth of CCGG/CCWGG) decreased significantly(P0.01), methylation sequencing depth decreased significantly(P0.01). The different functional components on the genome methylation level in two groups had statistically significant difference(P0.01). From the chromosome map, the distribution of difference in methylation loci on chromosomes was not uniform in two groups. Conclusion: The difference of whole genome methylation may be one of the mechanisms of the bystander effect of radiation injury rats.
