摘要:目的研究化淤软肝胶囊对四氯化碳致肝纤维化大鼠的抗肝纤维化作用及对肝组织中TGF-β_(1)表达的影响。方法将SD大鼠随机分为正常组、模型组、化淤软肝胶囊组(高、中、低剂量组)、秋水仙碱组,除正常组外,采用多因素方法建立四氯化碳致肝纤维化大鼠模型。连续给药4周后,检测各组大鼠血清的肝功能和肝纤维化指标,HE染色观察肝组织病理学改变,采用免疫组化SP法、RT-PCR法、Western blot法检测肝组织中TGF-β_(1)的含量和表达。结果肝纤维化大鼠肝损伤指标ALT、AST、DBIL和肝纤维化指标HA、LN、PC-Ⅲ、IVC的水平升高,而化淤软肝胶囊剂量组和秋水仙碱组明显降低,尤以高剂量显著,HE染色结果与之对应;化淤软肝胶囊能够抑制TGF-β_(1)蛋白和mRNA的表达,尤以高剂量明显。结论化淤软肝胶囊具有抗肝纤维化的作用,其作用机制可能与抑制TGF-β_(1)途径相关。
OBJECTIVE To study effects of Huayu Ruangan capsule(HYRG)on hepatic fibrosis induced by carbon tetrachloride and its effect on TGF-β_(1) in liver tissues.METHODS SD rats were randomly divided into normal group,model group,high dose group of HYRG(HYRG-H),middle dose group of HYRG(HYRG-M),low dose group of HYRG(HYRG-L)and Colchicine group(Col),and the carbon tetrachloride induced HF rat model was established by multi-factor method except normal group.After 4 weeks of administration,liver function and liver fibrosis indexes were detected.HE staining was used to observe liver histopathological changes,immunohistochemical SP,and RT-PCR and Western blot were used to detect the content and expression of TGF-β_(1) in liver tissue in each group of rats.RESULTS Levels of HF liver injury indexes including ALT,AST and DBIL,and liver fibrosis indexes including HA,LN,PC-Ⅲand IVC increased in rats.However,those indexes decreased significantly in HYRG groups and colchicine group,especially in HYRG-H group,where HE staining results correspond.HYRG could inhibit expression of TGF-β_(1) and its mRNA,especially for HYRG-H group.CONCLUSION HYRG inhibits the degree of liver fibrosis,and its mechanism is associated with inhibition of TGF-β_(1) pathway.
