摘要:目的采用HPLC指纹图谱技术研究45个不同产地、18批不同采收期、8批不同树龄所产枸杞子药材的差异性。方法采用Agilent HC-C18(4.6 mm×250 mm,5μm)色谱柱,流动相为0.1%冰乙酸溶液和甲醇,梯度洗脱;利用中药指纹图谱相似度评价软件对不同批次枸杞子药材进行相似度评价。结果采用指纹图谱相似度评价发现,产自宁夏中宁、甘肃靖远及新疆等地的枸杞子相似度最高,表明该区域枸杞子在化学成分方面具有一致性。甘肃河西地区、青海德令哈、内蒙古等地所产枸杞子与宁夏中宁枸杞子相比相似度较低。同一产地不同采收期枸杞子药材呈现出采收月份越近指纹图谱相似度越接近的趋势,且不同年份均呈此规律;不同树龄所产枸杞子药材随树龄越接近指纹图谱相似度越相近。结论本文研究结果为枸杞的药材质控及相关研究提供了借鉴和参考依据。
Objective To evaluate the difference of Fructus Lycii(FL)from 45 different producing areas,18 different harvesting periods and 8 different tree-age variations,by the HPLC fingerprint technique. Methods Agilent HC-C18(4.6 mm×250 mm,5 μm)chromatographic column was used,with the mobile phase of 0.1% glacial acetic acid-methanol in a gradient elution. The similarity evaluation software of fingerprint was used to evaluate the similarity of different batches of FL samples. Results The highest similarity was found in the FL samples from Zhongning city of Ningxia Autonomous Region(AR),Jingyuan county of Gansu Province and Xinjiang AR by the fingerprint similarity evaluation,indicating a consistency in the chemical composition of the FLs in these areas.By contraries,the similarity was lower in the FL samples from Hexi region of Gansu Province,Delingha of Qinghai Province,Inner Mongoria AR and other places. The FL samples harvested in different harvesting periods in the same place showed a tendency of changes in the similarity of fingerprints:the closer the harvesting months,the higher the similarity of fingerprints,and the same trend was seen in every one of the different years. The same was true of the case of FLs with different tree-age variations:the closer the tree ages,the higher the similarity of fingerprints. Conclusion The present results could be used for reference for the quality control of FLs and the related researches.
