摘要:以当归多糖为原料制备当归多糖锌复合物,研究锌离子对当归多糖在溶液中的自组装形貌及固体结构的影响,并且对当归多糖锌复合物的抗氧化活性进行评价。采用傅立叶变换红外光谱(FT-IR)、热重分析(TG)、X射线衍射(XRD)等手段对组装结果进行确认,通过扫描电子显微镜(SEM)、倒置荧光显微镜、透射电镜(TEM)、刚果红实验等手段对当归多糖锌复合物在溶液中的自组装形貌进行分析,通过对1,1-二苯基-2-苦苯肼自由基(DPPH)、2,2-联氮-二(3-乙基-苯并噻唑-6-磺酸)二铵盐自由基(ABTS)的清除率综合评价多糖锌复合物的抗氧化能力。实验结果表明:在溶液中,当归多糖锌能够自组装成囊泡结构,固态时形成有序的层状结构,扫描电镜和刚果红证实了Zn 2+掺入当归多糖高分子柔性链中,改变了多糖的构象。当浓度为1 g/L时,当归多糖锌复合物对DPPH自由基和ABTS自由基的清除率分别达到40.20%和86.18%。因此可以得出结论:锌离子的引入,改变了当归多糖在溶液中自组装的形貌,但是并没影响其固体层状结构。当归多糖锌复合物有较好的体外抗氧化活性。复合物在溶液中的形貌特征以及其抗氧化等实验结论可为开发兼具有当归多糖生物活性和抗氧化活性的药用补锌剂提供基础数据。
Angelica polysaccharide zinc complex based on angelica polysaccharide was prepared to study its self-assembled morphology and solid structure in solution and solid,the antioxidant activity of angelica polysaccharide zinc complex was evaluated at the same time.Fourier transform infrared spectroscopy,thermogravimetric analysis and X-ray diffraction were used to confirm the assembly results.The self-assembly morphology of angelica polysaccharide zinc complex in solution was analyzed by scanning electron microscope,inverted fluorescence microscope,transmission electron microscope and Congo red experiment.The antioxidant capacity of zinc polysaccharide complex was evaluated by means of DPPH and ABTS free radical.Results show that in solution angelica polysaccharide zinc was able to self-assemble into a vesicular structure and form an ordered lamellar structure in the solid state.Both scanning electron microscopy and Congo red experiments showed that Zn 2+incorporated into the polysaccharide polymer chain changed the conformation of the polysaccharide.When the concentration was 1 mg/mL,the scavenging rates of DPPH free radical and ABTS free radical were 40.20%and 86.18%,respectively.A conclusion can be got that the introduction of zinc ions changed the morphology of angelica polysaccharide in solution but did not affect its solid lamellar form.Angelica polysaccharide zinc complex had better antioxidant activity in vitro.The morphological characteristics of the complex in solution and the antioxidant data provided basic data for the development of medicinal zinc supplements with both biological and antioxidant activities of angelica sinensis polysaccharide.
