摘要:目的:观察槲皮素对阿霉素致大鼠肾小球硬化的干预作用。方法:采用单侧右肾切除术加两次尾静脉注射阿霉素建立肾小球硬化模型。将大鼠随机分为正常组、假手术组、模型组、槲皮素(25 mg·kg^(-1)·d^(-1)、50 mg·kg^(-1)·d^(-1)、100 mg·kg^(-1)·d^(-1))剂量组,总共干预8周。于槲皮素干预0、2、4、6、8周末检测24 h尿蛋白,实验结束后观察大鼠血生化、行HE染色观察肾组织病理并评估肾小球硬化程度,Western blot技术检测ZO-1、Nephrin、α-SMA以及FSP-1的表达。结果:与正常组和假手术组相比,模型组大鼠24 h尿蛋白量、尿素氮(BUN)、肌酐(Scr)、胆固醇(TC)、三酰甘油(TG)、肾小球硬化指数(CSI)均明显升高,肾小球α-SMA、FSP-1表达显著增加(P <0. 05),总蛋白(TP)、白蛋白(Alb)、肾小球ZO-1、Nephrin的表达均明显减少(P <0. 05)。与模型组相比,槲皮素干预组24 h尿蛋白量、BUN、Scr、TC、TG水平以及CSI呈剂量依赖性地降低,肾小球α-SMA、FSP-1的表达呈剂量依赖性地减少(P <0. 05),TP、Alb、ZO-1、Nephrin的表达呈剂量依赖性地增加(P <0. 05)。正常组与假手术组相比,各项指标差异无统计学意义。结论:槲皮素可以延缓肾小球硬化的发生。
Objective:To observe the effect of quercetin on adriamycin induced glomerular sclerosis in rats. Methods:The rat model of glomerular sclerosis was induced by tail intravenous injection of adriamycin twice.SD rats were randomly divided into normal group,sham operation group,model group and quercetin(25 mg·kg^-1 ·d^-1,50 mg·kg^-1 ·d^-1,100 mg·kg^-1 ·d^-1 )dose group,a total of eight weeks.The 24 h urinary protein was detected at 0,2,4,6,8 week after quercetin intervention.After the experiment,the serum creatinine was measured by automatic biochemistry analyzer,renal pathological changes were observed by HE staining,Glomerular sclerosis index(GSI) were caculated.The expressions of α-SMA,FSP-1,ZO-1 and Nephrin were examined by Western blot. Results:Compared with normal group and sham operation group,the results of 24 h urine protein,Scr,BUN,Cho,TG,CSI and the expressions of α-SMA and FSP-1 were higher in model group( P <0.05),TP,Alb,ZO-1 and Nephrin were lower in model group( P <0.05);Compared with model group,the results of 24 h urine protein,Scr,BUN,Cho,TG,CSI and the expressions of α-SMA and FSP-1 were lower in quercetin group ( P <0.05),TP,Alb,ZO-1 and Nephrin were higher in quercetin group( P < 0.05 );All indexes were no statistical significance between sham operation group and normal group ( P >0.05). Conclusion:Quercetin can ameliorate amycin-induced glomerular sclerosis.
