摘要:目的:优选红芪总黄酮超声提取的最优工艺并探讨提取物的抗氧化活性。方法:以总黄酮的得率及提取物的抗氧化活性为指标评价工艺,在单因素实验的基础上,运用正交试验法考察提取时间、超声功率、溶剂浓度三因素对红芪总黄酮得率及抗氧化活性的影响;紫外分光光度法测定提取物中总黄酮的含量,DPPH、O_2^-·法和ABTS 3种方法测定提取物的抗氧化活性。结果:红芪总黄酮超声提取的最优工艺为:70%乙醇,超声功率170 W,提取20 min,此工艺条件下提取的总黄酮具有较好体外抗氧化活性,高浓度的黄酮抗氧化活性相当于0.5 mg/mL维生素C,总黄酮对DPPH、O_2^-·和ABTS+的IC50分别为3.336 mg/mL、3.988 mg/mL、4.786 mg/mL。结论:经验证此工艺稳定可行,提取物抗氧化活性较传统的热回流法强,且呈一定的量效关系,可作为工业化大生产的参考。
To optimize the ultrasonic extraction technology of total flavonoids from Radix Hedysari and to explore the antioxidant activity of the extract. Methods:The effect of the yield of total flavonoids and the antioxidant activity of the extract were evaluated. On the basis of single factor experiment,the effects of three factors including extraction time,ultrasonic power and solvent concentration on the yield and antioxidant activity of the total flavonoids of Radix hedysari were investigated by orthogonal test. The antioxidant activities of the extracts were determined by three methods including DPPH,O2-and ABTS.Results:The optimum extraction process of ultrasonic extraction of total flavonoids from Radix Hedysari was:70% ethanol,power 170 W,20 min. The total flavonoids extracted under this condition had better antioxidant activity in vitro,the antioxidant activity of high concentration flavonoids was equivalent to 0.5 mg/mL VC,and the total flavonoids were 3.336 mg/m L,3.988 mg/mL4.786 mg/mL. to DPPH,O2-·and ABTS+ respectively. Conclusion:This process was stable and feasible,and the antioxidant activity of the extract was higher than that of the traditional heat reflux method,and it had a certain amount effect relationship,which could be used as a reference for industrial production.
