摘要:目的:观察当归补血汤含药血清对AngⅡ诱导的心肌细胞凋亡的影响,探讨其抑制心肌细胞凋亡的机制。方法:将心肌细胞分为4组:空白对照组、AngⅡ(10-7mol/L)组、缬沙坦(10-6mol/L)组、5g/kg当归补血汤1日2次的10%当归补血汤含药血清组。以Annexin V/PI流式细胞仪检测细胞凋亡率,透射电镜观察细胞线粒体超微结构,Western blotting测定凋亡相关蛋白Bax、Bcl-2表达变化。结果:10%当归补血汤含药血清干预后心肌细胞凋亡率明显低于AngⅡ(10-7mol/L)组,心肌细胞Bax蛋白表达显著低于AngⅡ组,Bcl-2蛋白表达显著高于AngⅡ组,Bax/Bcl-2比值显著低于AngⅡ组。结论:10%当归补血汤含药血清对AngⅡ诱导的心肌细胞凋亡有明显的抑制作用,其可能与减少心肌细胞Bax蛋白和增加Bcl-2蛋白表达相关。
Objective : To study effect of serum containing Dangguibuxue Decoction on the apoptosis of Ang II-induced cardiomyocytes,and discuss its mechanisms.Methods: The cultured cardiomyocytes were divided into 4 groups: control group,angⅡ(10-7mol / L) group,valsartan group(10-6mol / L) and 10% medicated serum group.Apoptosis rate was detected with Annexin V / PI flow cytometric analysis.Mitochondrial injury of the myocardium was studied with the electron microscope.Western Blotting was used to study the change of Bax protein,Bcl-2 protein.Results: The apoptosis rate of 10% medicated serum group was significantly lower than the AngII(10-7mol / L) group(P < 0.05).Bax protein of medicated serum group was obviously lower than the AngII group(P < 0.05).Bcl-2 protein of medicated serum group was significantly higher than the AngII group(P < 0.05) Bax / Bcl-2 was significantly lower than the AngII group(P < 0.05).Conclusion: 10% serum containing Dangguibuxue Decoction could inhibit the apoptosis of AngII-induced cardiomyocyte.It may be related with the reduce Bax protein and increase Bcl-2 protein.
