摘要:目的明确党参不定根悬浮培养的生长曲线,探究茉莉酸甲酯(MeJa)和壳聚糖(CTS)两种诱导子对不定根生长及多糖含量的影响。方法接种后不同天数测定党参不定根的干鲜重,向不定根悬浮培养体系中分别添加两种不同种类、不同浓度的诱导子。结果党参不定根生长曲线呈“S”型。茉莉酸甲酯的适宜添加浓度为150μmol/L,适宜诱导时间为8天;壳聚糖的适宜添加浓度为50 mg/L,适宜诱导时间为4天。结论在党参不定根的悬浮培养中,可在不定根培养的第15天加入适宜浓度的MeJa或CTS并诱导适宜时间,能显著提高党参不定根的多糖含量。
Objective To clarify the growth curve of the suspension culture of Codonopsis pilosula adventitious roots and to investigate the effects of two inducers,methyl jasmonate(MeJa)and chitosan(CTS),on the growth and polysaccharide content of the adventitious roots.Methods The dry and fresh weights of C.pilosula adventitious roots were measured at different days after inoculation.Two different types of inducers with varying concentrations were added to the adventitious root suspension culture system.Results The growth curve of C.pilosula adventitious roots showed an“S”shape.The optimal concentration of MeJa was 150μmol/L with a suitable induction time of 8 days;the optimal concentration of CTS was 50 mg/L with a suitable induction time of 4 days.Conclusions In the suspension culture of C.pilosula adventitious roots,adding an appropriate concentration of MeJa or CTS on the 15th day of culture and inducing for a suitable duration can significantly increase the polysaccharide content of the adventitious roots.
