摘要:目的研究加味当归贝母苦参丸对H22肝癌荷瘤小鼠血清白细胞介素(IL)-2、IL-4、γ-干扰素(IFN-γ)表达的影响。方法于昆明小鼠腋下注射接种H22肝癌细胞复制H22肝癌荷瘤小鼠模型,造模后将60只小鼠随机分为模型组、顺铂阳性对照组、加味当归贝母苦参丸高剂量组、加味当归贝母苦参丸低剂量组、加味当归贝母苦参丸高剂量联合顺铂组、加味当归贝母苦参丸低剂量联合顺铂组6组,每组10只。造模后第2天开始各组给予对应药物进行干预,连续治疗14 d后,摘除眼球取血并分离血清,采用ELISA法检测血清中IL-2,IL-4,IFN-γ的含量。结果与模型组及顺铂阳性对照组比较,其他各治疗组IL-4含量明显降低,IL-2,IFN-γ含量明显升高,差异均有统计学意义(P<0.05);模型组与顺铂阳性对照组各项指标比较,差异均无统计学意义(P>0.05);加味当归贝母苦参丸高、低剂量联合顺铂对各项指标的作用明显优于单纯加味当归贝母苦参丸高、低剂量,差异均有统计学意义(P<0.05)。结论加味当归贝母苦参丸可通过影响I L-2,IL-4,IFN-γ的表达调整机体免疫功能,从而起到抑瘤的作用。
: Objective To investigate the effects of modified Danggui (Angelica) Beimu (FritiUary) Kushen (Radix Sophora Flavescens) Wan on the expression of IL-2, IL-4 and IFN-γ in serum of H22 hepatoma-bearing mice. Methods Mice models were made by injecting H22 hepatoma cells into their armpits. 60 mice were ran- domly divided into model group, dichlorodiamineplatinmn (DDP) positive control group, high-, low-dosage treatment group (modified Danggui Beimu Kushen Wan only) , high-,low-dosage combination treatment group( modified Dang- gui Beimu Kushen Wan combined with DDP) , 10 mice for each. All groups were intervened with corresponding medicine on 2nd day after modeling, lasting 14 days. Blood was taken from eyeball and IL-2, IL-4 and IFN-γ in serum were detected to investigate the anti-tumor action and immunological mechanisms. Results Compared with model group and DDP positive control group, the content of IL-4 decreased, the contents of IL-2 and IFN-γ increased, the differences are significant ( P〈0.05 ). Comparison of all indexes between the model group and DDP positive control group has no any statistical significance (P〉0.05). Whatever the high- or low-dosage, effects in the combination treatment groups were superior to which in the single treatment groups, the differences had great significance (P〈0.05). Conclusion The modified Danggui Beimu Kushen Wan can regulate immunological mechanisms to inhibit tumor growth by adjusting the expression of IL-2, IL-4 and IFN-γ.
