摘要:以甘肃省7个春小麦品种为试验材料,探讨了接种方式、基因型、胚龄、植物生长调节剂等因素对幼胚培养中愈伤组织诱导及分化的影响。结果表明:供试春小麦品种出愈率不受盾片放置方向的影响,而盾片向上放置的分化率明显大于盾片向下放置的;春小麦幼胚离体培养的有效胚龄期为14~20d,最适宜的胚龄期为16d;基因型对愈伤组织诱导无影响,但对愈伤组织再生能力有极其重要的影响;随2,4-D浓度的增加春小麦幼胚分化率呈单峰曲线变化,且品种间差异很大,对大多数品种而言,适宜的2,4-D浓度为1~6mg/L。0.2mg/LNAA和0.2mg/LIAA能有效地诱导芽快速生根,生根率分别达到83.8%和55.1%。
Effects of inoculation direction, genotypes, embryo ages and plant growth regulators on callus induction and differentiation efficiency of in-vitro immature embryo of seven local spring wheat cultivars of Gansu were investigated in this research. The results indicated: inoculation direction had no significant effects on callus induction, but had significant effects on differentiation with up-shield inoculation having higher differentiation efficiency; the effective embryo ages for in-vitro immature embryo culture ranged from 14 days to 20 days, the optimal embryo age was 16 days; Callus induction had no significant difference among genotypes, but Callus differentiation and regeneration had great difference among genotypes. Differentiation rate presented single-curved trend with the increase of 2,4-D concentration, varying among genotypes, the suitable 2,4-D concentration for most of the cultivars was 1-6 mg/L, 0.2 mg/L NAA 和 0.2 mg/L IAA induced rooting promptly, with rooting rate up to 83.8% and 55.1%, respectively.
