摘要:目的:建立一套适用于甘草种子蛋白质提取及SDS-PAGE电泳的技术体系,为甘草种子纯度检测提供技术支撑。方法:对甘草种子蛋白质的8种提取方法进行了筛选,在此基础上进一步比较不同的分离胶浓度、不同料液比对甘草种子蛋白质SDS-PAGE电泳的影响,并采用该技术方法对乌拉尔甘草和胀果甘草种子进行鉴定。结果:巯基乙醇法和丙酮沉淀法是适宜的蛋白质提取方法,巯基乙醇法得到12条带,丙酮沉淀法得到9条蛋白质条带,且背景颜色较浅,适宜于进行种子纯度检测。10%分离胶和1∶10的料液比电泳效果较好。乌拉尔甘草和胀果甘草种子有蛋白质条带的差异。结论:建立了一种高质量的甘草种子蛋白质样品的制备方法及SDS-PAGE电泳技术,可用于甘草种子纯度检测。
Object:In order to test the purity of Glycyrrhiza uralensis Fisch.seeds,suitable technology systems of protein extraction and SDS-PAGE were studied.Method:Eight extraction methods for seeds protein extraction were tested.Choosing the best extraction method,the influence of different concentration of separation gels and different solid-liquid ratio on SDS-PAGE was studied.Results:Mercaptoethanol method and acetone precipitation method were suitable for protein extraction method,the method of mercaptoethanol got 12 bands,acetone precipitation got 9protein bands,and the background was distinct,which were suitable for seed purity testing.Electrophoresis effect of 10%separating gel and 1∶10solid-liquid ratio was fine.Seed-protein of Glycyrrhiza uralensis Fisch.and Glycyrrhiza inflate Bat.had differences in protein bands.Conclusion:a high quality of Glycyrrhiza uralensis seed-protein extracting method and SDS-PAGE electrophoresis technology were established,which can be used in seed purity testing.
