摘要:铁死亡是一种新型的细胞死亡方式,以细胞内铁过载和脂质过氧化物过度积累为特征,与多种骨科疾病的发生密切相关。骨质疏松是以骨密度降低为主要病理表现的慢性代谢性骨病,近年来随着研究的不断深入,发现铁死亡对骨代谢着重要的影响,而通过对铁死亡的调节以维持骨稳态,在骨质疏松的防治领域取得了一定的突破,展现出了良好的应用前景。该文阐述了铁死亡的发生机制和调节途径,梳理了铁死亡对骨代谢的调控机制,并总结了铁死亡在骨质疏松症发病及治疗中的研究进展,以期拓宽骨代谢的研究思路。
Ferroptosis is a novel mode of cell death characterized by intracellular iron overload and excessive accumulation of lipid peroxides,which is closely associated with the pathogenesis of many orthopedic diseases.Osteoporosis is a chronic metabolic bone disease characterized by decreased bone mineral density.In recent years,a large number of studies have been conducted to regulate bone homeostasis through ferroptosis to treat osteoporosis,and certain results have been obtained,showing a good research prospect.This article summarizes the mechanism and regulation pathway of ferroptosis,explores the correlation between ferroptosis and the pathological mechanism of osteoporosis,systematically elaborates on the effect of ferroptosis in osteoblasts,osteoclasts and bone marrow mesenchymal stem cells,and sorts out the therapeutic application of ferroptosis for osteoporosis in order to expand the therapeutic ideas of osteoporosis.
