摘要:目的该文旨在将院内栓剂制备为凝胶剂,以抗菌、止痒、抗炎等实验及网络药理学技术研究凝胶剂对阴道炎的治疗作用与可能机制。方法采用回流法提取复方有效成分,并以卡波姆基质等水溶性凝胶基质与药液进行胶化实验,对凝胶剂的性状、pH值、微生物限度及稳定性进行测定;采用网络药理学技术预测所得凝胶剂对阴道炎的可能作用机制;采用体内外抗菌、止痒、抗炎、抗过敏等试验考察凝胶剂对阴道炎的治疗作用。结果实验结果表明凝胶剂呈半透明状,在抗菌、止痒、抗炎、抗过敏等方面作用显著。网络药理学结果表明,此复方可作用于MAPK1、AKT1、JUN等关键靶点,参与营养水平反应、金属离子反应、酰胺结合,涉及脂质和动脉粥样硬化、PI3KAkt信号通路等。结论所制备的凝胶剂对阴道炎具有一定治疗作用,可为后续进一步机制研究奠定基础。
Objective The aim of this paper was to prepare in-hospital suppositories as gelata and to investigate the therapeutic effects and possible mechanisms of gelata on vaginitis using antibacterial,antipruritic and anti-inflammatory tests and network pharmacology techniques.Methods The active ingredients of the compound were extracted by reflux method and gelation experiments were carried out with water-soluble gelata matrix such as carbomer matrix and the drug solution.The properties,pH,microbial limit and stability of the gelata were determined.Results The results showed that the gelata were translucent,with significant antibacterial,antipruritic,anti-inflammatory and anti-allergic effects.The results of network pharmacology showed that this compound can act on key targets such as MAPK1,AKT1and JUN,participate in nutrient level reaction,metal ion reaction and amide binding,and involve lipid and atherosclerosis,PI3K-Akt signaling pathway,etc.Conclusion The prepared gelata has some therapeutic effects on vaginitis and may lay the foundation for further mechanistic studies.
