摘要:铁死亡是由不受控制的铁依赖性脂质过氧化所诱导的细胞程序性死亡的一种新形式,在众多疾病的发生发展中起着不可替代的作用。肌骨衰减疾病是指肌肉减少症(sarcopenia,SP)、骨质疏松症(osteoporosis,OP)以及肌少-骨质疏松症(osteosarcopenia,OS)等发生在肌肉骨骼系统的增龄代谢性疾病,具有较高的致残率和致死率,已成为全球范围内公共健康和社会经济的主要负担。近年来,铁死亡在肌骨衰减类疾病中的研究意义逐渐显现,并开展了较多研究。一般认为铁死亡是一把双刃剑,它既可以作为疾病发生的驱动因素,也可以利用它来逆转疾病表型。本综述以铁死亡为切入点,重点阐述铁死亡在肌骨衰减中的病理机制,并探讨铁死亡在肌骨衰减疾病中的潜在作用靶点,以期为肌骨衰减疾病管理开辟新的平台。
Ferroptosis is a new form of programmed cell death induced by uncontrolled iron dependent lipid peroxidation,which plays an irreplaceable role in the occurrence and development of many diseases.Musculoskeletal attenuation disease refers to aging metabolic diseases that occurs in the musculoskeletal system,such as sarcopenia(SP),osteoporosis(OP),and osteosarcopenia(OS),which lead to high disability and mortality rate and a major burden on public health and socio-economic worldwide.In recent years,the clinical significance of ferroptosis in musculoskeletal diseases has gradually emerged,and many studies have been conducted.It is generally believed that ferroptosis is a double-edged sword that can be used both as a driving force for disease occurrence and to reverse disease phenotypes.This review focuses on the pathological mechanism of ferroptosis in musculoskeletal decay and explores the potential targets of iron death in musculoskeletal decay diseases,to establish a new platform for management of musculoskeletal decay.
