摘要:目的探讨归芪定年方对D-半乳糖致衰老模型小鼠肝组织氧化损伤的影响。方法采用D-半乳糖诱导建立衰老模型小鼠,经归芪定年方灌胃干预3周后,应用ELISA检测肝组织中超氧化物岐化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活性及丙二醛(MDA)含量,HE染色检测肝组织病理学变化,Western blot和qPCR检测肝组织中TNF-α、IL-6、GPX1和IL^(-1)0水平。结果归芪定年方能有效改善衰老小鼠体征和肝细胞形态与结构,提高肝组织中SOD、GSH-Px(P<0.05)活性,降低MDA含量(P<0.05),促进GPX1、IL^(-1)0在肝组织中的表达和抑制TNF-α、IL-6的水平,差异均具有统计学意义(P<0.05)。结论归芪定年方通过调节SOD、GSH-Px活性及MDA、GPX1、IL^(-1)0、TNF-α和IL-6水平,有效改善D-半乳糖致衰老模型小鼠肝组织氧化。
Objective To explore the effect of Guiqi Dingnian Decoction on oxidative damage of liver tissue in D-galactose-induced aging model mice. Methods Aging model mice was established to be induced by D-galactose. After the intervention of Guiqi Dingnian Decoction for 3 weeks, the activities of superoxide dismutase(SOD), glutathione peroxidase(GSH-Px) and the content of malondialdehyde(MDA) existing in liver tissue were detected by ELISA, the histopathological changes of liver tissue were detected by HE staining, and the levels of TNF-α, IL-6, GPX1 and IL^(-1)0 in liver tissue were detected by Western blot and qPCR. Results Guiqi Dingnian Decoction could effectively improve the physical signs of the aging mice, morphology and structure of liver cells, increase the activity of SOD and GSH-Px(P<0.05), decrease the content of MDA(P<0.05), promote the expression of GPX1 and IL^(-1)0, and inhibit the levels of TNF-α and IL-6 in liver tissue. The differences were statistically significant(P<0.05). Conclusion Guiqi Dingnian Decoction can effectively improve the liver tissue oxidation of D-galactose-induced aging model mice by regulating the activities of SOD, GSH-Px and the levels of MDA, GPX1, IL^(-1)0, TNF-α and IL-6.
