摘要:目的分析甲状腺乳头状癌(PTC)中长链非编码RNA(lncRNA)的差异表达谱并预测其靶基因。方法用转录组测序技术对PTC及配对的甲状腺癌旁组织测序,以差异倍数(FC)作为标准筛选差异表达基因,并通过基因本体论(GO)和京都基因与基因组百科全书(KEGG)进行靶基因及相关信号通路的分析,以实时荧光定量-PCR法验证筛选的目的lncRNA。结果共筛选出差异表达的lncRNA 111个(P<0.05);差异表达的mRNA 29个(P<0.05),其中差异显著的lncRNA有18个,上调5个,下调13个(P<0.05)。差异表达靶基因主要参与维生素代谢、丝裂原活化蛋白激酶(MAPK)活性的激活和自噬等生物学过程。差异表达靶基因主要与以下几种传导途径相关:"焦粘连""cAMP信号通路""oxylocin信号通路"和"Wnt信号通路"。lnc01089-006和MSTRG.277779.1在甲状腺乳头状癌TPC-1细胞中表达下调。结论lnc01089-006和MSTRG.277779.1及其靶基因以及相关信号通路与甲状腺乳头状癌密切相关。
Objective To analyze the differential expression profile of long noncoding RNA(lncRNA) in papillary thyroid carcinoma(PTC) predict its target genes. Methods The transcriptome sequencing technologies was used for the thyroid tissue adjacent to carcinoma of PTC sequencing. The multiples differences fold change(FC) was as a standard screening differentially expressed genes. The target genes and related signaling pathways was analyzed through the Gene Ontology Gene Ontology(GO) and Kyoto Encyclopedia Gene and genome Kyoto Encyclopedia of Genes and Genomes(KEGG).The target lncRNA was preliminarily validated by quantitative real-time-PCR. Results A total of 111 differentially expressed lncRNAs were selected(P<0.05). There were 29 mRNA expressions(P<0.05), among which 18 lncRNAs were significantly different, 5 were up-regulated, and 13 were down-regulated(P<0.05). The differentially expressed target genes were mainly involved in vitamin metabolism, activation of mitogen-activated protein kinase(MAPK)activity, autophagy and other biological processes. The differentially expressed target genes were mainly related to signal transduction pathways such as"coke adhesion","cAMP signaling pathway","Oxylocin signaling pathway"and"Wnt signaling pathway". The lnc01089-006 and MSTRG. 277779. 1 were down-regulated in TPC-1 cells of thyroid papillary carcinoma. Conclusion Lnc01089-006 and MSTRG. 277779. 1,their target genes and related signaling pathways are closely related to PTC.
