摘要:目的:观察平胃胶囊对肝郁脾虚型功能性消化不良(functional dyspepsia,FD)模型大鼠促胃肠动力及血清、胃肠及下丘脑组织中胃动素(motilin,MTL)表达的影响。方法:将50只Wistar雄性大鼠行电极埋置术,术后随机选取8只大鼠作为正常组,剩余大鼠采用复合因素法建立FD大鼠模型,造模成功后随机分为:模型组、多潘立酮片10 mg/kg阳性组,平胃胶囊1.6 g/kg、3.2 g/kg、4.8 g/kg组,连续给药21天。检测各组大鼠造模及给药前后胃肠电活动,胃排空和肠推进率,采用ELISA法检测血清中MTL的含量,免疫组化SP法检测胃肠及下丘脑组织中MTL的分布和表达。结果:与正常对照组比较,模型组胃窦和十二指肠慢波振幅、快波振幅、快波发生率明显降低,胃排空、小肠推进率明显降低,血清胃动素含量降低,胃、十二指肠、下丘脑组织MTL表达明显下调。与模型组比较,平胃胶囊各剂量组明显增加胃窦慢波振幅、快波发生率,对十二指肠电活动有增加趋势,剂量依赖地增强胃排空率、小肠推进率,增加血清胃动素含量,上调胃、十二指肠和下丘脑MTL水平。结论:平胃胶囊能够促进肝郁脾虚型FD大鼠胃肠动力,其作用机理可能与上调血清、胃肠及下丘脑中MTL的表达有关。
Objective:To observe the effects of Pingwei capsule on the gastrointestinal motility,the expression of motilin(MTL)in serum,gastrointestinal and hypothalamic tissues in rats with Ganyupixu functional dyspepsia(FD).Methods:50 male Wistar rats were embedded with electrode.After the operation,8 rats were randomly selected as the control rats,the remaining rats were randomly divided into five groups after FD model were established:the model group,domperidone tablet group(10 mg/kg),Pingwei capsule groups(1.6 g/kg,3.2 g/kg,4.8 g/kg doses),rats in all groups were administered for 21 consecutive days.The gastrointestinal electrical activity,gastric emptying and intestinal propulsion rate were detected before and after the model establishment,before and after administration.The content of MTL in serum was detected by ELISA,the contents of MTL in gastrointestinal and hypothalamic tissues were detected by immunohistochemical method.Results:Compared with the control group,the slow wave amplitude,fast wave amplitude and fast wave num ratio of gastric antrum were decreased,the gastric emptying and the intestinal propulsion were significantly decreased,the serum level of MTL was decreased,the contents of MTL in gastrointestinal and hypothalamic tissues were down regulated.Compared with the model group,the slow wave amplitude and fast wave num ratio of gastric antrum in all drugs groups were increased,the duodenal electric activity was increased by Pingwei capsule.The gastric emptying and intestinal propulsion were improved,serum MTL was increased,the contents of MTL in gastrointestinal and hypothalamic tissues were up regulated.Conclusion:Pingwei capsule can promote the gastrointestinal motility in Ganyupixu FD rats,and its mechanism may be related to up regulating the levels of MLT in serum,gastrointestinal tract and hypothalamus.
