摘要:目的探讨IL-6/JAK2/STAT3信号通路对胃癌增殖的影响。方法建立MFC胃癌荷瘤小鼠模型,随机分为模型组、AG490阻断剂组、Stattic阻断剂组,每组12只,另选12只健康小鼠作为空白组。干预结束后取瘤体组织检测瘤体重量及计算抑瘤率;ELISA法检测小鼠血清中IL-6含量;免疫组化法检测瘤体组织中IL-6、JAK2、STAT3、c-Myc和Cyclin D1蛋白表达情况;蛋白免疫印迹法检测瘤体组织中IL-6、JAK2、p-JAK2、STAT3、p-STAT3、c-Myc和Cyclin D1蛋白表达情况;RT-qPCR法检测瘤体组织中IL-6、JAK2、STAT3、c-Myc和Cyclin D1 mRNA表达情况。结果与空白组比,模型组小鼠血清中IL-6含量升高(P<0.01);与模型组比,AG490阻断剂组和Stattic阻断剂组小鼠血清中IL-6含量,瘤体质量,瘤体组织中IL-6、p-JAK2、STAT3、p-STAT3、c-Myc和Cyclin D1的蛋白含量,IL-6、JAK2、STAT3、c-Myc和Cyclin D1 mRNA含量均下降(P<0.05,P<0.01)。结论阻断IL-6/JAK2/STAT3信号通路后,可影响下游靶蛋白的表达,进而抑制胃癌的增殖。
Objective To study the effect of IL-6/JAK2/STAT3 signaling pathway on the proliferation of gastric cancer.Method MFC gastric cancer tumor-bearing mice models were established and randomly divided into model group,AG490 blocker group,and Stattic blocker group,each with 12 mice,and another 12 healthy mice were selected as the blank group.After the intervention,the tumor tissue was taken to test the weight and calculate tumor inhibition rate;ELISA method to detect IL-6 content in mouse serum;Immunohistochemical method was used to detect IL-6,JAK2,STAT3,c-Myc and Cyclin D1 protein expression in tumor tissues;Western blot was used to detect IL-6,JAK2,p-JAK2,STAT3,p-STAT3,c-Myc and Cyclin D1 protein expression in tumor tissues;RT-qPCR method to detect IL-6,JAK2,STAT3,c-Myc and Cyclin D1 mRNA expression in tumor tissues.Result Compared with the control group,the IL-6 content in the serum of the model group was increased(P<0.01);Compared with the model group,serum IL-6 content,tumor mass,the protein content of IL-6,STAT3,p-JAK2,p-STAT3,c-Myc and Cyclin D1,and the mRNA content of IL-6,JAK2,STAT3,c-Myc and Cyclin D1 decreased(P<0.05,P<0.01).Conclusion Blocking the IL-6/JAK2/STAT3 signaling pathway can affect the expression of downstream target proteins,thereby inhibiting the proliferation of gastric cancer.
