摘要:目的:观察红芪多糖(HPS)对db/db小鼠心肌组织核转录因子(NF-κB p65)、基质金属蛋白酶-9(MMP-9)蛋白与mRNA表达的影响,探讨其防治心肌间质纤维化可能的作用机制.方法 :将60只雄性db/db小鼠随机分为5组:HPS高、中、低剂量组,罗格列酮组与模型组;12只雄性db/m小鼠为正常组.干预组小鼠连续灌胃8周,于干预前后每2周测空腹血糖、体质量各1次.第8周末心脏取血后,取心脏左室心肌组织,分别用于Masson染色、电镜观察心脏超微结构以及Western blotting、逆转录-免疫酶联反应(RT-PCR)法检测其NF-κB p65、MMP-9的蛋白与mRNA表达情况.结果:HPS干预8周后,与模型组比较:HPS高、中剂量组与罗格列酮组小鼠血糖下降明显(P<0.05),干预组小鼠体质量变化无明显差异(P>0.05);Masson染色可见亮绿色胶原纤维在心肌细胞间及血管周围明显增多且呈堆积状分布;电镜观察可见HPS高剂量组与罗格列酮组小鼠心肌组织内线粒体数量与破坏程度、糖原颗粒及脂滴沉积现象均有明显改善;Western blotting、RT-PCR检测结果显示,HPS高剂量组、罗格列酮组小鼠心肌组织中NF-κB p65、MMP-9的蛋白与mRNA表达量均明显降低(P<0.05).结论:HPS可降低db/db小鼠心肌组织中NF-κB p65、MMP-9蛋白与mRNA的表达水平,减轻其炎症反应状态,对心肌的间质纤维化病变具有一定的防治作用.
Aim: To observe the effect of HPS on the expression of NF-κB p65,MMP-9 protein and mRNA in myocardial tissue of db/db mice and to explore the possible mechanism of the prevention of HPS on myocardial fibrosis. Methods: Sixty male db/db mice were randomly divided into 5 groups: HPS high,middle,low dose of groups,rosiglitazone group and model group. Twelve male db/m mice were regarded as normal group. All the intervention groups were given continuous administration for 8 weeks.The fasting blood glucose and body weight were measured every 2 weeks. All the mice were sacrificed after 8 weeks of sdministration and their blood were pumped from heart. Taking part of the left heart tissue for Masson staining and the observation of electron microscopy. The rest of heart tissue for detection of the expression of NF-κB p65,MMP-9 protein and mRNA by the way of Western blotting and RT-PCR.Results: After the intervention of HPS for 8 weeks,the blood glucose of high and middle dose of HPS groups as well as rosiglitazone group decreased significantly compared to model group( P〈 0. 05),but the body weight were no significant difference among all the groups( P 〈 0. 05). The result of Masson staining showed that bright green collagen fibers increased in myocardial cells and blood vessels. The result of electron microscope showed that the deposition of collagen,the destruction of mitochonadria and the myocardial fibrosis were improved in both high dose of HPS group and rosiglitazone group. The expression of NF-κB p65,MMP-9 protein and mRNA decreased significantly in high dose of HPS group and rosiglitazone group( P 〈0. 05). Conclusion: HPS can reduce the expression of NF-κB p65,MMP-9protein and mRNA in the myocardial tissue of db/db mice and reduce the inflammatory response,which has a certain preventive and therapeutic effect on fibrosis of myocardial interstitial.
