摘要:目的研究乌梅丸对2,4,6-三硝基苯磺酸(TNBS)诱导的溃疡性结肠炎(UC)大鼠的作用机制。方法大鼠结肠灌注含50%乙醇的TNBS溶液复制UC模型。造模次日,按照体重将造模成功大鼠随机分为5组:模型组、阳性对照组和低、中、高3个剂量实验组,每组8只。另取8只正常大鼠作为正常组。阳性对照组大鼠灌胃0.5 g·kg^(-1) SASP,低、中、高3个剂量实验组分别灌胃7.2,14.4,28.8 g·kg^(-1)乌梅丸水提液,正常组与模型组灌胃生理盐水,均连续14 d。评价大鼠疾病活动指数(DAI),用酶联免疫吸附法检测结肠组织髓过氧化物酶(MPO)活性、白细胞介素6(IL-6)和肿瘤坏死因子α(TNFα)水平,用免疫印迹法检测结肠环氧酶2(COX-2)的表达。结果模型组、阳性对照组和低、中、高3个剂量实验组的DAI(第14天)分别为2.31±0.31,1.20±0.11,1.89±0.22,1.72±0.13和1.59±0.18分;这5组的MPO (U·mg^(-1))分别为0.95±0.04,0.34±0.02,0.77±0.02,0.47±0.04,0.44±0.04;这5组的IL-6 (pg·mg^(-1))分别为306.76±12.16,157.55±6.84,202.56±24.58,178.78±8.91,166.52±9.67;这5组的TNF-α(pg·mg^(-1))分别为450.11±72.87,288.79±7.59,346.37±9.53,318.09±24.00,303.89±12.96;这5组的COX-2的相对表达量分别为1.96±0.07,1.08±0.07,1.79±0.10,1.29±0.06,1.15±0.07。以上4个指标:模型组与正常组比较,差异均有统计学意义(P<0.01,P<0.05);3个剂量实验组与模型组比较,差异均有统计学意义(P<0.01,P<0.05)。结论乌梅丸对溃疡性结肠炎大鼠具有治疗作用,其作用机制可能与调节结肠IL-6、TNFα和COX-2表达有关。
Objective To study the effects and mechanism of Wumeiwan on the 2,4, 6-trinitrobenzenesulfonic acid(TNBS) induced ulcerative colitis in rats.Methods UC rats model were induced by 2,4,6-trinitrobenzenesulfonic acid(TNBS)/ethanol intra-colonic administration. Next day after modeling,successful modeling Wistar rats were randomly divided into 5 groups: model group, positive control group,low,middle and high doses experimental groups;another normal 8 rats was as normal group. Rats in normal group and model group were orally administered with normal saline,rats in positive control group were orally treated with sulfasalazine(0.5 g·kg^(-1)),while rats in experimental groups were orally administered with Wumeiwan decoctionat the dose of 7. 2,14. 4 and 28. 8 g·kg^(-1),respectively,for consecutive 14 d,1 times a day. The disease activity index( DAI) were scored. The levels of myeloperoxidase( MPO) activity,interleukin-6( IL-6) and tumor necrosis factor α( TNF-α) in the colon were determined by enzyme-linked immunosorbent assy. The level of cyclooxygenase-2( COX-2) in the colon were determined by Western blot. Results The levels of DAI( at fourteenth day) in normal group,positive control group,and low,middle and high doses experimental groups were 2. 31 ± 0. 31,1. 20 ± 0. 11,1. 89 ± 0. 22,1. 72 ± 0. 13 and 1. 59 ± 0. 18,respectively;the MPO content in the 5 groups were( 0. 95 ± 0. 04),( 0. 34 ± 0. 02),( 0. 77 ± 0. 02),( 0. 47 ± 0. 04),( 0. 44 ± 0. 04) U·mg^(-1),respectively;the IL-6 content in the 5 groups were( 306. 76 ± 12. 16),( 157. 55 ± 6. 84),( 202. 56 ± 24. 58),( 178. 78 ± 8. 91),and( 166. 52 ± 9. 67)pg·mg^(-1),respectively;the TNFα content in the 5 groups were( 1. 96 ± 0. 07),( 1. 08 ± 0. 07),( 1. 15 ± 0. 07),( 1. 29 ± 0. 06),and( 1. 79 ± 0. 10) pg·mg^(-1),respectively. Significant difference of the factors were found between model group and normal group( all P < 0. 05,P < 0. 01);and significant difference of the factors were also found between the three experimental groups and model group( P < 0. 05,P < 0. 01). Conclusion Wumeiwan has therapeutic effect on rats with UC,the mechanisms may be related to adjusted the levels of IL-6,TNFα and COX-2 in colon.
