摘要:目的探讨桃红四物汤加味防治膝关节骨性关节炎的机制。方法选用Wistar大鼠50只,随机分为空白组、模型组、四妙汤组、桃红四物汤组(简称桃红组)和吲哚美辛组,后4组进行膝关节骨性关节炎模型制作,用药6周后处死动物,免疫组化法检测MMP-1及TIMP-1。结果桃红组和吲哚美辛组大鼠与模型组比较,MMP-1的表达均有不同程度的降低(P<0.05),而TIMP-1的表达均有不同程度的升高(P<0.05)。结论桃红四物汤加味能有效地抑制Hulth法诱导的大鼠膝关节骨性关节炎软骨中MMP-1的表达,促进关节炎软骨中TIMP-1的表达。四妙汤加味对模型大鼠膝关节骨性关节炎软骨中MMP-1,TIMP-1的表达则无明显作用。
Objective To inquire into the mechanism of modified Taohong Siwu Tang on preventing and treating KOA. Methods 50 Wistar rats were randomly divided into bland group, model group, Simiao Tang group, Taohong Siwu Tang group and indomethacin group. Except bland group, rats of other group were made into KOA models. Six weeks later, the rats were executed,and MMP-1 and TIMP-1 were determined by Immunohistochemical Method. Results Compared with model group, the expression of MMP-1 was obviously decrease in Taohong Siwu group and indomethacin group( P 〈 0.05 ) , and the expression of TIMP-1 was obviously increase( P 〈 0.05). Conclusion Taohong Siwu Tang can effectively inhibit the expression of MMP-1 in articular cartilage of the rats with KOA induced by Hulth method, and promote the expression of MMP-1. While Simiao Tang have no obvious effect on the expression of MMP-1 and TIMP-1.
