摘要:目的探讨地龙蛋白对糖尿病勃起功能障碍(DMED)大鼠的干预作用及其可能机制。方法SPF级雄性SD大鼠60只,其中空白组10只,余下50只通过腹腔注射链脲佐菌素(STZ)构建糖尿病大鼠模型,8周后使用阿朴吗啡(APO)筛选符合DMED大鼠,成模大鼠随机分为模型组、西地拉非组、地龙蛋白低剂量组、地龙蛋白中剂量组、地龙蛋白高剂量组,药物干预4周后,测定ICP/MAP评估大鼠勃起功能。检测血清中超氧化物歧化酶(SOD)、丙二醛(MDA)、睾酮(T)含量,苏木精-伊红(HE)染色观察睾丸、阴茎组织结构的改变;免疫组化法(IHC)检测睾丸组织核因子E2相关因子2(Nrf2)、核因子κB(NF-κB)表达。结果与空白组相比,模型组大鼠血糖(4.78±0.89 vs 27.65±8.29,P<0.001)、血清MDA(2.85±1.02 vs 6.01±0.45,P<0.01)、睾丸组织NF-κB(0.285±0.014 vs 0.413±0.018,P<0.01)明显升高;体重(513.33±38.94 vs 289.33±48.96,P<0.001)、ICP/MAP(79.02±4.33 vs 23.20±1.92,P<0.01)、血清SOD(83.72±2.60 vs 38.52±1.43,P<0.01)、T(100.14±6.65 vs 41.51±2.88,P<0.01)、睾丸组织Nrf2(0.195±0.004 vs 0.108±0.002,P<0.01)显著下降。与模型组相比,各治疗组大鼠血糖、体重无统计学意义,血清MDA、睾丸组织NF-κB明显下降,ICP/MAP、血清SOD、T、睾丸组织Nrf2含量显著增加。HE结果显示,空白组大鼠睾丸、阴茎形态结构正常,模型组大鼠睾丸、阴茎组织损伤严重,各治疗组损伤均有所改善。结论地龙蛋白具有改善糖尿病大鼠勃起功能作用,其机制可能与抑制睾丸氧化应激、炎症反应,增加血清T含量有关。
Objective To investigate the intervention effect of earthworm protein on erectile dysfunction in diabetic rats and its possible mechanism.Methods Total of 60 SPF-grade male SD rats were used in the study,Ten of them were taken as the blank group,and the other 50 rats were injected with streptozotocin to form the diabetic rat models.Eight weeks later,apomorphine was used to screen the rats with diabetic erectile dysfunction.Rats with diabetic erectile dysfunction were further randomly divided into the model group,the sidirafil group,and low-dose,middle-and high-dose earthworm protein groups.After4 weeks of drug intervention,ICP/MAP was measured to evaluate erectile function of rats.The levels of serum SOD,MDA and T in serum were detected,and the changes of testis and penis tissue structures were observed after HE staining;the expressions of Nrf2 and NF-κB in testis tissue were detected by immunohistochemistry(IHC).Results Compared with those of the blank group,blood glucose level(4.78±0.89 vs 27.65±8.29,P<0.001),serum MDA level(2.85±1.02 vs 6.01±0.45,P<0.01)and testicular NF-κB expression(0.285±0.014)in the model group vs 0.413±0.018,P<0.01)were all significantly increased,whereas body weight(513.33±38.94 vs 289.33±48.96,P<0.001),ICP/MAP(79.02±4.33 vs 23.20±1.92,P<0.01),serum SOD level(83.72)±2.60 vs 38.52±1.43,P<0.01),T level(100.14±6.65 vs 41.51±2.88,P<0.01),testicular Nrf2 expression(0.195±0.004 vs 0.108±0.002,P<0.01)were decreased significantly.Compared with those of the model group,the level of serum MDA and the expression of testicular tissue NF-κB were significantly decreased in each treatment group,and the ICP/MAP,levels of serum SOD and T,and the expression of testis tissue Nrf2 were remarkably increased,but little changes were found in blood glucose level and body weight.HE staining results showed that the testis and penis of rats in the blank group were normal in shape and structure,while the testis and penis of rats in the model group were severely injured,and the damage was improved in each treatment group.Conclusion Earthworm protein can improve erectile function of diabetic rats,and its mechanism may be related to inhibiting testicular oxidative stress,inflammatory response and increasing serum T content.
