摘要:目的研究藏药湿生扁蕾不同溶剂提取物对四氯化碳致小鼠急性肝损伤的保护作用。方法将54只雄性小鼠随机分为空白组、模型组、联苯双酯对照组、湿生扁蕾水提取物组、湿生扁蕾醇提取物组和湿生扁蕾醇水提取物组6组,每组9只。各组分别灌服生理盐水,联苯双酯溶液,湿生扁蕾水提取物、乙醇提取物、乙醇和水提取物,灌胃7 d后,除空白组外,其余各组均灌服1.5%四氯化碳花生油20 mL/kg造模。24 h后经眼眶静脉丛取血,检测小鼠血清谷丙转氨酶(ALT)、谷草转氨酶(AST)活性及肝组织中丙二醛(MDA)含量、谷胱甘肽过氧化物酶(GSH-Px)活性,并观察小鼠肝脏病理学改变。结果与空白组比较,模型组小鼠血清ALT,AST活性及肝组织中MDA含量明显升高,肝组织中GSH-Px活性明显降低,差异均有统计学意义(P<0.01);与模型组比较,湿生扁蕾不同溶剂提取物组及联苯双酯对照组小鼠血清ALT,AST活性及肝组织中MDA含量明显降低,肝组织中GSH-Px活性明显升高,差异均有统计学意义(P<0.05或P<0.01),其中以湿生扁蕾醇水提取物组作用最为显著(P<0.05)。肝脏病理学观察结果显示,造模后模型组肝损伤程度严重,湿生扁蕾不同溶剂提取物组肝脏病理损伤程度明显减轻。结论藏药湿生扁蕾不同溶剂提取物可减轻四氯化碳诱导的小鼠急性肝损伤,改善小鼠肝功能,作用机制可能与其清除自由基、保护肝细胞膜结构和功能的完整性、提高肝细胞内的GSH-Px活性、增强机体抗氧化能力有关。
Objective To study the protective effect of extracts from Gentianopsis paludosa( Tibetan medicine) in different solvents on mice with acute liver injury caused by carbon tetrachloride( CCl_4). Methods 54 male mice were randomly divided into blank group,model group,bifendate control group,Gentianopsis paludosa water-extract group,Gentianopsis paludosa alcohol-extract group and Gentianopsis paludosa alcohol-water-extract group,9 mice in each; all groups were administered with saline,bifendate,water extract,alcohol extract and alcoholwater extract for 7 days respectively; then after that,except the blank group,the rest groups were fed with CCl_4 peanut oil for modeling. 24 hours later,blood were taken from the orbital venous plexus to detect serum ALT and AST,HE staining was used to observe the histopathological changes of mice liver,MDA and GSH-Px in liver homogenate were tested. Results Compared with blank group,the activity of serum ALT and AST,the content of MDA in model group increased obviously,and the level of GSH-Px decreased,the differences were significant( P〈0.01); compared with model group,other groups intervened with extracts of Gentianopsis paludosa in different solvents reduced the contents of ALT,AST and MDA clearly and enhanced the activity of GSH-Px,there were significant differences( P〈0.05 or P〈0.01),and alcohol-water extract got the most notable effect( P〈0.05). Pathological observation showed severe liver injury in model group,while the injury were relieved in other extracts groups. Conclusion Extracts of Gentianopsis paludosa can relieve the liver injury and improve liver function with the possible mechanism in eliminating free adicals,protecting the integrity of liver cells membrane structures and function,enchancing the activity of GSH-Px and antioxidative capacity.
