摘要:目的探讨^(12)C^(6+)束辐射大鼠的肺-心-肝-脾旁效应损伤及归芪益元膏对其的防治作用及机制。方法选取健康雄性Wistar大鼠90只,随机分为正常对照组(NC组)、单纯辐射组(SR组)、归芪益元膏组(GO组),每组30只。NC组给予2ml生理盐水灌胃,辐射剂量0Gy,SR组给予2ml生理盐水灌胃,辐射剂量8Gy,GO组给予11.83g/kg归芪益元膏灌胃,辐射剂量8Gy,大鼠连续灌胃给药7d,行^(12)C^(6+)束辐射右侧肺部造模。SR组和GO组大鼠造模后48h处死,取心、肝及脾组织。采用比色法测定丙二醛(MDA)、还原型谷胱甘肽(GSH)、谷胱甘肽过氧化物酶(GSH-Px)、超氧化物歧化酶(SOD)含量;ELISA法检测DNA甲基化率;免疫组织化学法检测Dnmt1、Dnmt3a、Dnmt3b表达。结果与NC组比较,SR组大鼠SOD、GSH、GSH-Px降低(P<0.01),MDA升高(P<0.01);DNA甲基化水平降低(P<0.01);Dnmt1、Dnmt3a、Dnmt3b表达升高(P<0.01)。与SR组比较,GO组大鼠SOD、GSH、GSH-Px升高(P<0.01),MDA下降(P<0.01);DNA甲基化水平升高(P<0.01);Dnmt1、Dnmt3a、Dnmt3b表达降低(P<0.01)。三组Dnmt1、Dnmt3a、Dnmt3b蛋白均有表达,且分布在心肌细胞、肝细胞、脾脏白髓外周B细胞胞质内。NC组颜色为浅褐色-褐色,呈弱阳性表达;SR组颜色为褐色-棕色,呈强阳性表达;GO组颜色为浅褐色-棕褐色,呈中等度阳性表达。结论归芪益元膏能减轻^(12)C^(6+)束辐射造成的旁效应,其机制与改善氧化应激反应及DNA甲基化水平有关。
Objective To investigate the bystander effect injury to lung-heart-liver-spleen caused by 12 C6+ beam radiation, and explore the prevention and treatment effect of Guiqiyiyuan ointment on the injur y and its mechanism. Methods Ninety healthy male Wistar rats were randomly divided into 3 groups: NC group(normal control, normal saline 2 ml/kg, n=30), SR group(simple radiation, 8 Gy, 2 ml/kg, n=30), GO group(Guiqiyiyuan ointment 11.83 g/kg, radiation, 8 Gy, n=30). All the rats received intragastric administration for 7 days. The right side of the lung was modeled by 12 C6+ beam radiation. After modeling, the rats were killed at 48 h. The heart, liver and spleen were taken. The malonaldehyde(MDA), glutathione(GSH), glutathione peroxidase(GSH-Px), superoxide dismutase(SOD) contents were measured by colorimetry, DNA methylation rate was assayed by ELISA, and the expressions of Dnmt1, Dnmt3 a and Dnmt3 b were detected by immunohistochemistry. Results Compared with NC group, the contents of SOD, GSH and GSH-Px decreased(P〈0.01), MDA increased(P〈0.01), the level of DNA methylation decreased(P〈0.01), and the expressions of Dnmt1, Dnmt3 a and Dnmt3 b increased in SR group(P〈0.01). Compared with SR group, the contents of SOD, GSH and GSH-Px increased(P〈0.01), MDA decreased(P〈0.01), the level of DNA methylation increased(P〈0.01), and the expressions of Dnmt1, Dnmt3 a and Dnmt3 b decreased in GO group(P〈0.01). Dnmt1, Dnmt3 a and Dnmt3 b proteins were expressed in the cytoplasm of myocardial cells, hepatocytes and peripheral B cells of the white pulp in spleen, in all the groups. The color of NC group was light brown-brown, showing a weak positive expression. The color of SR group was brown-brown, showing a strong positive expression. The color of GO group was light brown-tan, showing a moderate positive expression. Conclusion The Guiqiyiyuan ointment can reduce the bystander effect caused by the 12 C6+ beam radiation, and its mechanism is related to improving the oxidative stress reaction and the level of DNA methylation.
