摘要:目的观察集束化疗法对心搏骤停(CA)模型兔心肺复苏(CPR)后血流动力学指标、磷脂酰肌醇3-激酶(PI3K)/蛋白激酶B(Akt)/糖原合成酶(GSK)3-β蛋白变化的影响,探讨其心脏保护的作用机制。方法将24只日本大耳白随机分为假手术组、肾上腺素组、亚低温组、集束化治疗组,每组6只。采用体外电击法复制兔CPR模型。于不同时间点动态监测血流动力学指标、PI3K/Akt/GSK3-β蛋白表达。结果与假手术组比较,肾上腺素组、亚低温组和集束化治疗组复苏成功后各时间点血流动力学指标、PI3K/Akt/GSK3-β蛋白水平均明显改善,其中以集束化治疗组效果最显著(均P<0.01)。结论集束化疗法对CPR后兔心肌有明显保护作用,其机制与改善血流动力学、激活PI3K/Akt/GSK3-β信号通路有关。
Objective To observe the effect of cluster chemotherapy on hemodynamic parameters and PI3K/AKT/Gsk3-β protein after cardiopulmonary resuscitation in rabbits with cardiac arrest, and to explore the mechanism of cardioprotection. Methods Twenty-four Japanese white rabbits were randomly divided into sham operation, adrenaline, mild hypothermia and cluster treatment groups, with 6 rabbits in each group. CPR rabbit model was reconstructed by external electric shock. The changes of hemodynamic parameters and PI3K/AKT/Gsk3-β protein were dynamically monitored at different time points. Results Compared with those of sham operation group, the hemodynamic indexes and PI3K / AKT / Gsk3-β protein levels of adrenaline group, mild hypothermia group and cluster treatment group were significantly improved at each time point after resuscitation, especially in cluster treatment group ( P <0.01). Conclusions Cluster chemotherapy has obvious protective effect on rabbit myocardium after CPR. The mechanism is related to improving hemodynamics and activating PI3K/Akt/GSK3-β signaling pathway.
