摘要:目的通过观察大剂量当归水煎液对氢溴酸东莨菪碱所致小鼠记忆障碍的保护作用,探讨当归防治老年性痴呆的作用机理。方法 (1)采用氢溴酸东莨菪碱腹腔注射诱发小鼠记忆障碍,而后以大剂量当归进行干预,采用水迷宫法、跳台法及避暗法进行行为学能力测试,并测定脑组织中乙酰胆碱酯酶(TchE)和胆碱乙酰转移酶(ChAT)的活性。结果大剂量当归和小剂量当归治疗组均能减少在水迷宫测试、跳台测试、避暗测试中的错误次数减少,缩短潜伏时间;升高脑组织匀浆ChAT活性、降低AchE活性,其中以大剂量当归组效果最为明显。结论大剂量当归具有防治老年性痴呆的作用,其机理可能与改善其学习记忆能力,提高脑组织中胆碱能神经递质的活性,保护神经细胞有关。
Objective This study is to explore the mechanisms of water extraction of Angelica with large doses on the protective effect of scopolamine hydrobromide-induced memory impairment in mice and treatment of senile dementia.Methods The experimental model of memory impairment was induced with Hydrobromide scopolamine alkali intraperitoneal injection,and then intervention of large doses of angelica also was carried out on the model mice.After that,water maze test,step down test and darkness avoidance test were conducted to test behavior proficiency,and to test acetylcholinesterase(TchE) activity and choline acetyltransferase enzyme(ChAT) activity of brain tissue.Results large doses of angelica and small doses of drug treatment group can reduce the error number and shorten latency in the water maze test,step down test and darkness avoidance test;and could increase the brain homogenate ChAT activity,lower AchE activity.especially the effects of large doses of angelica group were obviously.Conclusion Large doses of angelica has the function of preventing and treating senile dementia,and the mechanisms of that may be associated with the mprovement of learning and memory ability,and the improvement of activities of cholinergic neurotransmitters in the brain tissue to protect nerve cells.
