摘要：目的评价二十五味儿茶凝胶治疗小鼠慢性变应性接触性皮炎(ACD)的疗效。方法采用2,4-二硝基氯苯(DNCB)诱导小鼠背部变应性接触性皮炎。60只小鼠随机分为正常对照组、模型组、丁酸氢化可的松(尤卓尔)组、二十五味儿茶凝胶高剂量、中剂量、小剂量组,每组10只,于致敏后给药,观察各组小鼠第4次激发24h后皮损炎症程度、组织病理切片、对小鼠免疫器官的影响及血清中IL-17和TNF-α水平的变化。结果二十五味儿茶凝胶中、大剂量能有效减轻慢性ACD小鼠引起的免疫器官肿大,减轻皮损炎症程度,减少组织炎症细胞数量,调节ACD组小鼠血清中TNF-α和IL-17水平。结论二十五味儿茶凝胶对小鼠慢性ACD具有明显的治疗作用,其作用机制可能与其降低TNF-α、IL-17,调节了Th1、Th17型细胞因子平衡,通过多途径抑制自身免疫反应的发生有关。
Objective To evaluate the efficacy of catechu gelatin of twenty-five ingredients in the treatment of chronic allergic contact dermatitis（ACD）in mice. Methods 2,4-dinitro-chlorobenzene（the DNCB）induced allergic contact dermatitis in mice back. Sixty mice were randomly divided into six groups,including control group,model group,hydrocortisone butyrate ointment treatment group,low,medium and high doses of catechu gelatin of twenty - five medicine group. 10 mice were treated after sensitization in each group. The changes of the degree of skin lesions,histopathological sections,the immune organs in mice and the changes of serum IL-17 and TNF-α levels after 24 hours by the fourth stimulation were observed. Results Medium and high dose of catechu gelatin of twenty-five ingredients can effectively reduce chronic allergic contact dermatitis in mice caused by immune organ enlargement,reduce the degree of inflammation of the lesion,reduce the number of inflammatory cells in the organization and regulate TNF-α and IL-17 in serum of ACD mice group. Conclusion Catechu gelatin of twenty-five ingredients has significant therapeutic effect on chronic allergic contact dermatitis in mice. Its mechanism relates to down-regulating TNF-α and IL-17,maintaining the balance of Th1 and Th17 cytokines,and inhibiting the occurrence of autoimmune response through multiple channels.
