摘要:目的:探究有利于诱导半夏试管珠芽形成与试管苗生长的Mn^(2+)、Zn^(2+)浓度。方法:以半夏试管苗叶柄作外植体,1/2 MS培养基为基本培养基,改变培养基中Mn^(2+)与Zn^(2+)浓度,观察统计其对半夏试管珠芽形成及试管苗生长的影响。结果:通过形态指标统计与淀粉粒数量对比,发现试管珠芽分别在Mn^(2+)浓度为100μmol/L、Zn^(2+)浓度为120μmol/L时诱导珠芽形态最佳,单珠芽诱导率最高且愈伤诱导率最低,植株生长最健壮,试管珠芽组织切片顶端分生组织下方所含淀粉粒数量最多,分布最均匀。结论:适宜浓度Mn^(2+)、较高浓度Zn^(2+)有利于半夏试管珠芽形成、试管苗生长及淀粉粒积累,且淀粉粒积累与植株生长呈正相关。
Objective:To explore the concentrations of Mn^(2+)and Zn^(2+)which were beneficial to the formation of bulbils and the growth of test-tube seedlings of Pinellia ternata.Methods:The petioles of P.ternata test-tube seedlings were used as explants,and 1/2 MS medium was used as the basic medium.The concentrations of Mn^(2+)and Zn^(2+)in the medium were changed,and the effects on the formation of bulbils and the growth of test-tube seedlings were observed and statistically analyzed.Results:Through the comparison of morphological index statistics and the number of starch grains,it was found that when the concentrations of Mn^(2+)and Zn^(2+)were 100μmol/L and 120μmol/L respectively,the induction of test-tube bulbils showed the best results.The morphology of the bulbils was the best,the induction rate of single bulbils was the highest and the callus induction rate was the lowest,and the plant growth was the most robust.The number of starch grains contained under the apical meristem of the test-tube bulbil tissue section was the largest and the distribution was the most uniform.Conclusion:The appropriate concentration of Mn^(2+)and higher concentration of Zn^(2+)are beneficial to the formation of bulbils,the growth of test-tube seedlings and the accumulation of starch granules,and the accumulation of starch granules is positively correlated with the growth of plants.
