摘要:目的采用电感耦合等离子体质谱法(ICP-MS)及原子荧光光谱法(AFS)测定不同生长期党参地上、地下部位中28种无机元素的含量及动态特征。方法样品经微波消解后,采用ICP-MS法测定27种无机元素的含量,AFS法测定Hg元素的含量。并结合聚类热图、相关性、主成分及正交偏最小二乘-判别分析等方法分析其规律。结果28种无机元素在实验浓度范围的线性关系均良好(r^(2)≥0.9973),检测限为1×10^(-4)~5.063μg·L^(-1);精密度、重复性、稳定性试验的RSD均小于4%,平均加样回收率为86.51%~110.88%,RSD为0.16%~6.65%(n=6)。党参地上部位元素的含量在10~11月最高,地下部位的含量在7~11月先降后升。K、Ca、Mg为引起地上、地下部位差异的主要元素。结论所用方法操作简便、灵敏度高,能全面分析党参中无机元素的动态特征,为党参药材的质量控制提供了参考。
OBJECTIVE To determine the contents and dynamic characteristics of 28 inorganic elements in the aboveground and underground parts of Codonopsis pilosula at different growth stages using inductively coupled plasma mass spectrometry(ICP-MS)and atomic fluorescence spectrometry(AFS).METHODS After microwave digestion,the contents of 27 inorganic elements were determined by ICP-MS,and the content of Hg element was determined by AFS.Combined with clustering heat map,correlation,principal components and orthogonal partial least squares-discriminant analysis,the pattern was analyzed.RESULTS The linearity of the 28 inorganic elements was good(r^(2)≥0.9973),and the detection limits ranged from 1×10^(-4)μg·L^(-1)to 5.063μg·L^(-1).The RSDs of the precision,reproducibility,and stability tests were less than 4%.The average spiked recoveries ranged from 86.51%to 110.88%,with the RSDs varying from 0.16%to 6.65%(n=6).The element content of aboveground parts of C.pilosula reached the highest between October and November,while the element content in the underground parts decreased first and then increased from July to November.K,Ca,and Mg were responsible for the differences between the two parts of C.pilosula.CONCLUSION The method is simple to operate and highly sensitive,and can comprehensively analyze the dynamic characteristics of inorganic elements in C.pilosula,providing a reference for the quality control of C.pilosula.
