摘要:目的研究野菊花提取物对慢性支气管炎(CB)大鼠肺功能及血清、肺组织中超氧化物歧化酶(SOD)、丙二醛(MDA)、一氧化氮(NO)含量表达的影响,以期探讨其治疗CB的部分疗效机制。方法制备野菊花提取物;SPF级Wistar大鼠随机分为空白对照组,模型组,喘息灵对照组,野菊花提取物低剂量组、中剂量组、高剂量组;以气管内注入脂多糖建立CB大鼠模型,药物干预后,检测各组大鼠肺功能,血清及肺组织匀浆中SOD活性、MDA及NO的含量。结果模型组大鼠较空白对照组大鼠呼吸频率加快,最大呼气幅度减小,最大吸气幅度加深,模型组大鼠血清、肺组织中MDA、NO含量较空白对照组明显升高,SOD活力显著下降(P<0.05),而经野菊花提取物治疗后,各组大鼠呼吸功能明显改善,且能显著提高SOD水平,降低MDA、NO含量(P<0.05)。结论野菊花提取物具有明显改善CB大鼠呼吸功能及自由基代谢的作用。
Objective To study the effect of the extract of Chrysanthemum Indicum on the protein expression of superoxide dismutase(SOD),malondialdehyde(MDA) and nitric oxide(NO) content of serum,lung tissue in chronic bronchitis(CB) rats in order to find out the effect mechanism of its treatment of CB.Methods Extract of Chrysanthemum indicum was prepared.SPF Wistar rats were randomly divided into blank control,CB model,chuanxiling group,low,middle and high dose groups of Chrysanthemum Indicum extract.CB rats were established with endotracheal injection of lipopolysaccharide.After drug intervention,the expression of SOD,MDA,NO content of rats' serum and lung tissue and function of lung were detected.Results Compared with the blank control group,the respiratory frequency of CB rats sped,the biggest expiratory amplitude and maximal inspiratory amplitude of them were decreased and deepened;the MDA,NO content of CB rats' serum,lung tissue were increased significantly,but SOD content was decreased significantly(P〈0.05).After treated with Chrysanthemum Indicum extract,the abnormal rising of MDA,NO were reduced(P〈0.05) and the level of SOD was improved significantly;Meanwhile,CB rat's respiratory function was improved obviously.Conclusions Chrysanthemum indicum extract has obviously improved CB rats' respiratory function and metabolic function of free radicals.
